葉開溫2006-07-262018-07-062006-07-262018-07-062004-07-31http://ntur.lib.ntu.edu.tw//handle/246246/17465為了探討文心蘭(Gower Ramsey)在花梗發育過程中,養分儲存與開花品質及 產期相關功能性基因體分析, 利用cDNA 微陣列和扣減式EST 文庫研究了 在花梗發育初期(花梗長度2-4cm)假球莖中特異表現的基因: 在扣減式EST 文庫中選出14 個與碳水化合物,包括蔗糖、澱粉、甘露糖及果膠,代謝相關基 因以北方墨漬法分析表現,其中13 個於假球莖中會大量表現,推測出文心蘭假 球莖在花序發育過程中可能先以甘露聚糖後以澱粉的形式貯存累積。在扣減式 EST 文庫中也發現class III peroxidase(POXs)會大量表現,並從文心蘭cDNA 文庫 中,找出了11 條POXs 基因。現正著手將這些POXs 基因轉植到阿拉伯芥與文 心蘭中,進而研究它們於花序發育生理功能的作用與影響。The investigation of nutrient storage, floral quality and floral time in Oncidium Gower Ramsey pseudobulb during the development of flower stalk were carried out. At the initiation of flower stalk (length 2-4 cM), more than 1000 pseudobulb specific expressed genes were discovered using cDNA microarray and subtractive EST library; The Northern blot analysis of fourteen cDNA in the subtractive EST library related to carbohydrate metabolism group confirm that thirteen cDNA have higher expression level in pseudobulb, suggesting mucilage may be formed first and then transferred to starch during the development of flower stalk. We also found that class III peroxidase (POXs) were highly expressed in the subtractive EST library and obtained eleven POXs genes form Oncidium cDNA library.
