2014-08-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/694567摘要:粉蝨傳播的番茄黃化捲葉病是最具破壞性的新興植物病害之一,全球許多作物生產皆受到其嚴重威脅。番茄黃化捲葉病可能導致高達 100% 的產量損失,可見其對農業經濟的損害相當大。這個疾病的發生與一群親緣相近的病毒有關,這群病毒稱為番茄黃化捲葉病毒 (屬於 genus Begomovirus, family Geminiviridae)。儘管番茄黃化捲葉病極具經濟上的重要性,但是我們對於病毒與病媒昆蟲交互作用的知識仍顯得不足。缺乏這些相關基礎知識對防治蟲媒疾病 (insect-borne disease) 是非常大的缺陷。因為要防治蟲媒疾病必先對付其病媒昆蟲,缺乏對病毒與病媒昆蟲交互作用之了解,使我們無法有效地控制昆蟲媒介的疾病。我們的遠程目標是要了解植物病毒和病媒昆蟲之間的交互作用。這個研究計畫的主要目的是研究番茄黃化捲葉病毒和煙草粉蝨 (Bemisia tabaci) 的交互作用。我們將多方面運用分子生物學、組織學、電生理學與生態學等實驗技術來達成此計畫目標。我們將探討番茄黃化捲葉病毒的感染對煙草粉蝨生存、發育、壽命和繁殖力之影響。再利用取食電位圖譜 (electrical penetration graph) 技術來分析此類病毒的感染對煙草粉蝨探測與取食行為之影響。最後,我們將採用免疫組織學與分子生物學工具,檢驗此類病毒對病媒昆蟲的致病性。詳細了解粉蝨和番茄黃化捲葉病毒之間的交互作用,或許可以協助我們發展新穎的防治策略,保護農作物免於受到此類病毒的危害。<br> Abstract: Whitefly-transmitted tomato yellow leaf curl disease is one of the destructive emerging diseases that threaten many crops worldwide. Tomato yellow leaf curl disease may cause yield losses of as much as 100% in tomato; therefore, the disease impairs the agriculture economic greatly. The disease is associated with a group of phylogenetically related viruses named tomato yellow leaf curl viruses (genus Begomovirus, family Geminiviridae). Despite the economic importance of tomato yellow leaf curl disease, little information for the virus-vector interaction is known. Lacking information about virus-vector interaction impairs the efficiency of insect-borne disease control because the insect vector is the best target for the disease system. Our long-term goal is to understand the interactions between plant viruses and their insect vectors. The main objective of the proposed research is to study the interactions between tomato yellow leaf curl viruses and Bemisia tabaci using a variety of techniques including molecular biology, histology, electro-physiology, and ecology. We will examine the effect of virus infection on the survival, development, longevity, and fecundity of B. tabaci. We will further analyze the impact of virus infection on the probing and feeding behavior of B. tabaci by electrical penetration graph. Finally, we will employ immunohistology and a molecular biological tool to examine the pathogenicity of these viruses on their insect vector.
