2016-08-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/694274摘要:Neutrohpil gelatinase-associated lipocalin(NGAL)為隸屬於lipocalin家族之大小約25 kDa分泌性醣蛋白,以monomer、dimer及NGAL/MMP-9複合物以上3種形式存在。許多人醫研究皆指出,腎臟受損後NGAL會於短時間大量表現於血液及尿液中,因此NGAL於早期預測腎衰竭之角色也備受重視。目前犬隻NGAL已被確定可以成為預測急性腎損傷及評估腎衰竭臨床進展的指標。雖然貓的NGAL序列已被發表,但在貓的腎病中所扮演的角色則尚未有人探討。本實驗室已於今年製備出貓NGAL多株抗體,此研究主要為利用此貓NGAL多株抗體,建立ELISA系統偵測臨床上所收集的貓隻血清及尿液樣本中NGAL濃度,並以Western blotting分析貓尿中NGAL分子型態,再由統計分析貓尿中NGAL濃度是否和犬隻及人類相同和腎病具有相關性,另外再探討其型態是否會受及其他泌尿道疾病所影響,希望藉此可對於NGAL於貓腎病中的表現有更深入的瞭解,以提升貓NGAL於臨床應用上之準確性。<br> Abstract: Neutrophil gelatinase-associated lipocalin (NGAL), a 25 kDa glycoprotein existing as monomer, homodimer, or NGAL/MMP-9 complex, was found highly accumulated in human and mouse proximal tubule during acute tubular necrosis, thus regarded as an early predictor of acute kidney injury (AKI) and related to the progression of chronic kidney diseases (CKD). Urine NGAL had also been found to serve as a sensitive and specific biomarker for the prediction of renal injury in dogs. Although the sequence of feline NGAL (F-NGAL) had been identified; however, the role of NGAL in feline has not yet been studied. The aim of the present study was to use polyclonal F-NGAL antibody in-house generated to set up an ELISA system, by which the serum and urine NGAL concentration in feline with different kinds of kidney diseases will be measured. Furthermore, molecular forms of NGAL in the clinic specimens of feline urine and serum will be determined by Western blot analysis.
