2005-01-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/684159摘要:因應外國稻米之開放進口及分子標記之最新發展,目前白米DNA之鑑定技術有賴進一步之提昇及實用化,以確保白米交易秩序,建立市場白米品質之區隔性及維持競爭力。本計畫擬以不同水稻品種Waxy基因之5' 非轉錄區; 3' 非轉錄區及外子 5至10之PCR所得DNA片段定序比對,建立單核&#33527;酸多型性資料庫。同時開發新的SSR分子標記,並持續擴增現有水稻新品種DNA指紋資料庫。本計畫亦朝向將此技術優化,建立標準程序之multiplexSSR或SNP,以期提高效率、精確性並達到技術移轉之實用化程度,以利本國稻米產業之發展與經營。<br> Abstract: In response of the import of foreign rice grains and the newly development of DNA markers, there is a demand to improve the current DNA fingerprinting technology of rice grains characterization. Therefore, the trade order, grain quality,compartmentation and competition ability of local rice market are able to maintain and continous to sustain. The aim of this study is to construct a SNP database by alignment of the sequence from the PCR products (5' UTR, 3'UTR and exon 5 to 10) of waxy gene from different rice cultivars. Meanwhile, the screening of novely polymorphic SSR markers and broad the DNA fingerprinting database of recent breeded rice cultivars will be carried out. The final goal for this project is to optimize the above results, develope multiple SNP or SSR and set up a standard operation procedure to further increase the efficiency, accuracy of current rice grain DNA fingerprinting technique.
