2006-01-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/684219摘要:為確保我國食米市場之有效管理及國產米之品質,本計畫擬發展食米單一核甘酸多型性(SNP)分子鑑識技術,朝向晶片分析、快速自動化檢測技術之開發。目前先以參與水稻澱粉合成及分解之基因共26個,利用Tilling (targete induced local lesion in genomics)及amplicon melting curve analysis之方法,尋找在不同稻米品種中單一核&#33527;酸差異之多型性,建立實際可用之SNP指紋分析資料庫。並將以allele-specific PCR等方法進行不同稻米品種之SNP genotyping。本計畫建立之分析技術及標準程序,將可配合現今之美、日等國、提供比過去已知方法更敏感,準確性亦更佳之食米品種檢驗平臺,將來亦具產學合作之開發價值。<br> Abstract: In order to maintain the effective management of local rice market and rice grain quality, this project focus on developing SNP genotyping technology for different rice cultivars. We also try to set up a long term goal in future for high-through put and automatic analysis of rice genotyping with SNP chip or SNP microarray.. Currently, we use both tilling and amplicon melting curve analysis methods and include more than 26 genes involved in biosynthesis and degradation of starch to screen for SNPs in various rice. By this way we hope to build up a SNP database for practical application. At last, an allele specific PCR method will also be used in real to test the effectivness of SNP genotyping in different rice. The technology set up in this project is compatible to those developed in Japan or USA and would provide a platform for much more sensitive, accurate and rapid rice cultivars characterization.
