2013-10-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/656966摘要:台灣紅豆杉是經國際自然保護聯盟瀕危物種名錄(IUCN red list)中,列為稀有及瀕臨絕滅的原生裸子植物,也是重要的林木經濟資源。在台灣紅豆杉族群,近年來更因為森林破壞、人類製作家具或提煉藥物需求,更加急遽消失之中。因此,了解其野生族群的遺傳變異程度,釐清其與亞洲鄰近地區紅豆杉的親緣,來提供政府保育方針建議,是刻不容緩的議題。本計畫將調查台灣紅豆杉幾個殘存野外族群及林試所保存的野生種源族群,利用4個葉綠體及1個核DNA 分子標記,來分析台灣紅豆杉野生及種源族群之遺傳歧異度概況,鑑定出哪些族群具有較高遺傳多樣性,並估算這些殘存族群是否正遭遇瓶頸效應,族群數量是否正在減少之中,藉此提供保育單位規劃參考。並藉由和中國大陸及菲律賓紅豆杉屬近緣種,比對親緣地理關係,以釐清台灣紅豆杉的起源及正確的分類命名,以了解其與東亞紅豆杉不同種群間是否具有遺傳獨特性。<br> Abstract: Taiwanese yew (Taxus summatrana) is listed crtically endangered in IUCN red list of gynmnosperm worldwide. In Taiwan it is even under great threatening that many populations are diminishing due to human activities on deforestation, or because of extensive anthropogenic usages such as medicine and furniture making. Therefore it is extremely important to assess the genetic diversity among individuals of these extant Taxus populations to determine which populations really need recovery action. There is also a need to elucidate the phylogeographic relationship between Taiwanese yew and adjacent Philippines and Chinese yews to clarify its phylo-taxonomic position. This project aims to analyze the diversity in Taiwanese yew utilizing four chloroplast genetic markers (trnL-F, trnH-psbA,petG-trnP, atpI-atpH)with one nuclear genetic marker(ITS).With these data, we could estimate the extent of genetic variation among each investigated population, identify populations that preserve ancestral or unique haplotypes, monitor the demography of these population to understand whether some of them are in bottle neck or declining. In gathering these info, we could propose a government conservation policy or plan.In addition, in resolving the phylogeographic relationship of Taiwanese yew to other yews from adjacent regions, we could trace the origin of Taiwanese yew, its proper taxonomic position and its ancestral colonization route into Taiwan.
