2005-01-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/657087摘要:本年度將進行完整的貓兒眼睛草屬植物的野外採樣,並萃取其genomic DNA。目 標在選殖出貓兒眼睛草屬植物小分子量熱休克蛋白基因家族中的葉綠體型可能成 員,其主要方法首先利用小分子量熱休克蛋白基因特定引子進行PCR的選殖,所 得到的片段再經由克隆並定序,若無法順利鑑定出這個基因片段,則將採取替代 的釣取方法如在建構小型的基因庫(genomic library)之後,利用南方轉漬的方法選 殖具有小分子量熱休克蛋白基因雜合訊號的菌株,再進一步鑑定其序列。之後並 希望能進一步確認這些相關基因所扮演的功能,找出可能的基因調控路徑圖。具 體目標為:1. 完整的貓兒眼睛草屬植物的野外採樣,並萃取其genomic DNA。 2. 選殖出貓兒眼睛草屬植物小分子量熱休克蛋白基因家族中葉綠體型基因。 3. 培養研究人才,鼓勵學生參與研究。<br> Abstract: In the facing year, we will collect all the necessary plant materials from three species of Chrysosplenium in Taiwan. Genomic DNAs will be extracted and subjected for PCR. The main goal is to identify the plastid small heat shock protein homologues from Chrysosplenium. PCR will be carried out using degenerate primers designed from known sequences in GenBank. The PCR products will be cloned and sequenced. If we cannot obtain the correct sequences, alternative approaches will be used that will require southern blotting using sHSP probe on a sub-genomic library. The specific objectives are: 1. Complete the material collection and extract the genomic DNA; 2. Identify the plastid small heat shock protein from selected Chrysosplenium species; 3.
