2013-02-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/684881摘要:豬是人類最早飼養的動物之一,而豬隻之馴化改變了野豬遺傳、形態與性能。近年來,許多研究著重於馴化事件對於豬隻性狀之影響,同時篩選出與性狀功能相關之單點核&#33527;酸多型性(single nucleotide polymorphism, SNP)取代位點。家豬之免疫力強弱受到人類選拔導致某些特定基因之基因型與野豬不同。利用不同豬種體染色體分子標記之多型性探討上述性狀改變,成為現今研究之趨勢。免疫反應為豬隻非常重要的對抗外源抗原性狀。位於Toll-like receptor 基因上許多之單一核&#33527;酸多型性(SNP),被認為與豬隻免疫力增加有關。此外,亦有研究指出Fucosyltransferase (FUT1)基因多型性與仔豬離乳後下痢有關。但過去研究沒有比較馴化前後這些基因之序列變化,也沒有利用近親品種做為研究模式增加研究的可靠性。臺灣野豬與李宋豬基因序列的變化,提供一個重要研究模式。去年,我們著重於免疫與仔豬下痢相關基因在不同豬種間單一核&#33527;酸多型性之研究。藉由比較亞洲家豬、歐洲家豬、與李宋豬與臺灣野豬,篩選出上述性狀相關基因之SNP多型性。延續去年計畫,今年預計建立李宋豬對腸毒素(enterotoxin)敏感性檢測、lipopolysaccharide (LPS)處理李宋豬後血液與免疫相關蛋白濃度變化檢測。期中,完成李宋豬之Toll-like receptor基因與LPS處理後免疫相關之不同基因型分群、完成李宋豬fucosyltransferase 基因與仔豬下痢相關之不同基因型分群。期末,完成李宋豬之免疫相關基因Toll-like receptor之基因型與抗病力關係之分析、完成李宋豬之抗下痢相關基因fucosyltransferase之基因型與抗病力關係之分析。此研究將可以將李宋豬開發成一個適合做為探討與免疫相關的實驗豬模式。<br> Abstract: Pig (Sus scrofa) is one of earliest domestic animals. The genetic, morphology and performance were changed after domestication. In recent years, many studies are focusing on the performance changes after domestication event of livestock and screening the single nucleotide polymorphism (SNP) on the andidate genes. The strength of immune-related genes in response to pathogen is altered after domestication. The SNPs on particular genes has been selecting by different molecular markers located on different pig breeds’ chromosomes. The performance of immune in response to ectopic pathogen is a very important trait for livestock industry. The SNP in Toll-like receptor gene and Fucosyltransferase gene is important in immune response ability and defense to diarrhea induced by pathogens, respectively. Either Toll-like receptor gene or Fucosyltransferase gene was done by comparison the SNP in the two genes in between wild pig and domestic pig. The sequences differences between Formosan wild pig and Lee-sung pig proved an excellent model for selection SNP in the genes related to immune response. Last year, the SNP of Toll-like receptor gene and Fucosyltransferase gene among Asian, European, Lee-sung and Formosan wild pig were identified. To extend the research, we will examine the sensitivities of Lee-sung pigs after enterotoxin or lipopolysaccharide treatments by the sign of diarrhea and immune-related proteins in blood. In the midterm report, the Lee-sung pigs will be separated to different groups by their genotyping of Toll-like receptor gene or Fucosyltransferase gene. Finally, the their genotyping (SNP) and the performances related to Toll-like receptor gene or Fucosyltransferase gene will be study and analysis.
