2012-04-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/679945摘要:骨質疏鬆症的原因很多,特別是老年人為好發階段,是造成骨質耗損的結果。本研究的目的是以李宋迷你豬為實驗動物,先以抽血檢測血清中的Vitamin D,骨鹼性磷酸酶(BAP)和C-端交聯端肽的I型膠原(CTX)在正常時的含量,同時以功能性的核磁共震儀(MR)檢測腰椎L2-L3-L4之椎骨動脈的血流速度和骨質,另外以BMD(骨礦物質密度)儀檢測正常值,另外從豬抽取骨髓幹細胞,做為第一階段工作。然後將豬做卵巢截除術,如女人的停經,等待骨質舒鬆症的產生。此期間將骨髓幹細胞經由細胞流速儀篩選純正的骨髓間葉幹細胞(MSC),在培養皿內誘導為骨母細胞,並大量培養以供治療之用,此為第二階段。經由一段長時間的等待,再抽血檢測血清中的Vitamin D,骨鹼性磷酸酶(BAP)和C-端交聯端肽的I型膠原(CTX)的含量之變化,以便進行第二次的功能性的核磁共震儀(MR)檢測腰椎L2-L3-L4之椎骨動脈的血流速度之減緩和骨質變鬆,另外以BMD(骨礦物質密度)儀檢測密度值的下降,藉以確定骨質疏鬆症的造成,此為第三階段。第四階段是將大量的骨母細胞注射入回豬之骨髓腔內,並等待一段時間,再抽血檢測血清中的Vitamin D,骨鹼性磷酸酶(BAP)和C-端交聯端肽的I型膠原(CTX)的含量之恢復,做為骨質恢復之參考,然後再進行第三次的功能性的核磁共震儀(MR)檢測腰椎L2-L3-L4之椎骨動脈的血流速度之恢復和骨質之恢復正常,另外以BMD(骨礦物質密度)儀檢測密度值的恢復,藉以確定骨質疏鬆症的治療成功。這樣的研究成果可供人類治療骨質疏鬆症的參考,而另一部份的對照組可以繼續供相關代謝性疾病的研究,把臺大育成的李宋迷你豬推向國際舞台。<br> Abstract: The osteoporosis caused by many reasons, especially results in the bone resorption. The purpose of this study is to induce the osteoblast from the bone marrow mesenchymal stem cell and in order to use them for bone regeneration by perfusion into the bone marrow cavity of porcine osteoporosis. First, In the preliminary experiment, the bone marrow mesenchymal stem cell was collected from Porcine and purify cultured preparation and then created an animal model of osteoporosis with Lee-Sung mini pigs under ovariectomy. Next induce the bone marrow mesenchymal stem cells into the osteoblast and perfusion of the osteoblast or the osteoblast contained green fluorescence gene into the bone marrow cavity after the porcine osteoporosis induced. Comparison of the control group and experimental group used Bone mineral density (BMD) Measurements and MR Measurements. In addition to performVitamin D assay, bone alkaline phosphatase (BAP), and C-terminal cross-linked telopeptide of type I collagen (CTX) assay. The control group of porcine osteoporosis should used for another research of medical metabolism in the future. The mesenchymal stem cells derived from bone marrow used very popularly, if the induced osteoblast from the mesenchymal stem cells could regenerate the bone formation in the osteoporosis patients, we think this is the novel treatment in the osteoporosis medical field.
