2007-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/711481摘要:脂肪細胞除了可貯存能量,並能分泌許多具有細胞激素效能的產物。脂肪細胞結合素(adiponectin)屬於此等細胞激素,首先於1995被Scherer等人選殖,它只在脂肪細胞表現並可被分泌至血液循環。此激素可增進小鼠肝臟及肌肉對葡萄糖的利用與脂肪酸的氧化,所以可減輕二型糖尿病的症狀。我們已選殖豬的adiponectin及其受體(AdipoR1和AdipoR2)的全長cDNA序列,並發現該等受體基因的表現遍佈各種組織,顯示adiponectin可至全身發揮其功能。但限食對此二受體在豬隻脂肪組織的表現調控不同,顯示二者功能調控機制或有不同。目前對此二受體功能及調控尚未釐清,所以本計畫將以下述研究來探討豬脂肪細胞結合素及其受體的功能表現調控機制。 一、 選殖基因、產製胜&#32957;製造抗體,以研究脂肪細胞結合素及其受體的蛋白質表現(現已完成胜&#32957;的產製)。 二、 限食對豬隻各組織中AdipoR1和R2表現之影響,以瞭解營養狀態的效應。 三、 以RNAi抑制AdipoR1或AdipoR2的表現,探討此二基因的個別功能及其對下游基因調控機制。 四、 產製重組豬的adiponectin(已完成表現載體構築),以研究此蛋白質的功能。 五、 選殖此二受體基因的啟動子序列,以研究營養分及內泌素對此等基因的調控。 六、 產製轉基因小鼠以研究豬的AdipoR1和AdipoR2對體內營養分利用的影響。 七、 產製轉基因豬,以探討此等受體之功能,發展減少豬隻背脂堆積的新途徑。 <br> Abstract: Besides being utilized as energy storage cells, adipocytes can secret several cytokine-like substances. Adiponectin is one of the substances secreted by adipocytes and was discovered by Scherer in 1995. Treatments of adiponectin increase glucose utilization and fatty acid oxidation in the liver and muscle of mice. Therefore, it can also alleviate the symptom of type II diabetes. Our preliminary experiment showed that a feed restriction treatment in pigs regulated the two adiponectin receptors in adipose tissue differently, indicating different regulation mechanisms involved in the process. Currently, the regulation of the expression of these two receptors is not clear in animals. Therefore, seven experiments will be carried out to study the regulation mechanisms of the functional expression of porcine adiponectin and adiponectin receptors. 1. To generate adiponectin peptide for antibody production in order to study the expression of adiponectin. We have had the peptide synthesized and are currently inducing the antibody production. 2. To study the effect of feed restriction on the expression of AdipoR1 and R2 mRNA in porcine tissues. 3. To study the function of AdipoR1 and R2 genes through interference RNA techniques. 4. To synthesize recombinant adiponectin for utilization in studying the function of adiponectin. 5. To clone the promoter regions of the two receptors in order to study the nutritional and hormonal regulation on the expression of these two genes. 6. To generate transgenic mice for studying the functions of these two receptors in vivo. 7.
