2006-01-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/661103摘要:此計畫將與另兩子計畫(黃文理博士及張孟基博士之計畫)採分工合作之方式,探討水稻對環境逆境反應之機制。本實驗室主要負責持續性的針對由T-DNA插入突變庫(由中央研究院余淑美博士團隊所建立)所篩選出之非生物性(乾旱、滲透、高鹽及溫度)逆境相關突變株,以PCR之方法得到T-DNA所標的基因之完整 cDNA或啟動子序列,並分析此些基因於臺農67水稻各發育期之表現、組織表現專一性及其在各環境逆境下之表現反應。此外,將以基因轉殖方法產生overexpression轉殖系進行各基因之功能分析。根據研究結果,進一步探討水稻對不同環境逆境反應機制之相關性。此部分之研究結果將提供給此整合計畫之各個參與實驗室以做為其進一步之基因應用的基礎。另一方面,於本實驗室先前之相關研究中,已篩選出數個高鹽(NaCl)逆境相關之水稻T-DNA插入突變株,並已由RT-PCR 方式得到此些目標基因(例如: molybdenum cofactor sulfurase),且由初步之分析已知此些基因之表現明顯的受NaCl所調控。因而在此計畫中,亦將接續探討此些基因受NaCl 調控之訊息傳導途徑,藉此以探討水稻對高鹽逆境之調控機制,<br> Abstract: The goal of this project is to realize the mechanisms of abiotic-stress (e.g. salt, drought and cold stress) tolerance in rice plants. To achieve this, we will cooperate with the other laboratories (Dr. Wen-Li Hung and Dr. Men-Chi Chang’s lab.) to identify the genes that contribute to the phenotype of hypertolerance/ hypersensitivity to abiotic stresses from the T-DNA inserted rice mutant library generated by Dr. Su-May Yu. In the network system of this consortium project, we are in charge of using molecular approaches to isolate and characterize the target genes or promoters of the T-DNA inserted stress-responsive mutants. Following, the gene expressions would be analyzed under abiotic stresses. Furthermore, the function of the interested genes could be identified in overexpression transgenic plants. The results would be provided to other cooperated laboratory for further research or application. Moreover, since there are several salt-stress responsive genes have been isolated and characterized in our previous project, we will continuously to study the signal transduction pathway of salt stress to regulate the salt-responsive genes.
