2005-07-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/661376摘要:翠菊黃萎病菌質體(aster yellows phytoplasma group, AY group)寄主範圍相當廣泛,感染植物後導致作物品質不良嚴重影響商品價值。台灣至今尚無此一病害之危害報告,目前國內部分作物採用國外進口之活植物苗為繁殖材料,如菊花、聖誕紅扦插苗,此類活植物苗之輸出國多屬於翠菊黃萎病菌質體之疫區,因此必須擬定精確快速之防檢疫策略,防範由植物活體之輸入導致病原菌入侵及蔓延,而造成對我國農業之衝擊。過去對於此病害之檢疫多藉由病徵觀察及顯微鏡技術,其精確靈敏度及時效性明顯不足;近來分子生物學的發達,學者多利用聚合酵素連鎖反應(PCR)進行此病原菌偵測。本計畫擬針對翠菊黃萎病菌質體發展篩檢健康活植物苗之偵測技術,以防止此一病原菌入侵;並同時進行感染薔薇科、菊科與葡萄科等作物之X-disease病原菌質體之偵檢工作(台灣至今亦無此一病害之報導),以達到無疏漏之植物病原菌質體檢疫功效。本計畫應用本研究室所研發之植物菌質體廣用型引子對f1/ r1(林及林,1998),以及Lee等人在1994年所發表之植物菌質體廣用型引子對R16F2/ R2(Lee et al., 1994)進行<br> Abstract: Aster yellows is caused by Aster yellows (AY) phytoplasmas and with wide host ranges. This pathogen has not been reported in Taiwan yet. Some seedlings of flowers are imported from pest infestation area of AY such as the Netherlands and the U. S. Therefore, this pathogen may be introduced by imported seedlings from those countries. In the past, detection of the AY phytoplasmas is mainly by symptom and by microscopic examination of phloem tissues. However, different phytoplasma types can not be differentiated by those methods. Furthermore, the accuracies and sensitivities of the above-mentioned methods are not enough and they are time-consuming. PCR technology has been developed for detecting phytoplasmas. This project is to develop a PCR-based detection system for detecting AY phytoplasma in imported seedlings.
