2008-04-092024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/664294摘要:一、目的 本研究擬進行紫芝醱酵液多醣之分離與純化,並以小鼠巨噬細胞模式,探討活性 多醣對巨噬細胞誘發免疫反應之調節機制、訊息傳遞途徑及功能驗證,以期確立 台灣紫芝生物活性多醣之免疫調節活性機轉,開發台灣紫芝於保健食品上之用途 為目的。。 二、材料與方法 1.菌種:擬採用台大生化科技學系許瑞祥教授所提供之台灣紫芝菌株。 2.以已建立之最適化醱酵生產條件於5公升小型醱酵槽生產台灣紫芝胞外多醣。 3.探討紫芝活性多醣刺激樹突細胞成熟之活性。 4.探討紫芝活性多醣活化小鼠巨噬細胞之訊息傳遞途徑,特別針對絲裂原活化蛋 白激&#37238;(MAP Kinases)及NF-κB之活化進行探討。 三、預期之結果 本研究若能順利達到確立台灣紫芝醱酵多醣之免疫調節活性及其功能評估,並應 用於生產台灣紫芝生物活性代謝產物之目的,則對國內在開發食藥用菇類之研究 及應用方面將有所貢獻。<br> Abstract: 1.Specific Aims The aims of this project are to purify the immunomodulating polysaccharides from the submerged culture of Ganoderma formosanum and to investigate the mechanisms of immunomodulation and signal transduction pathways in murine macrophages. These results will provide information for developing the G. formosanum polysaccharides into functional health food. 2.Materials and Methods (1)Microorganisms: G. formosanum provided by Prof. Hseu, National Taiwan university, will be used in this study. (2)Immunomodulating polysaccharides will be produced by culturing G. formosanum in a 5-liter jar fermentor. (3)Maturation of dendritic cells will be evaluated upon stimulation with G. formosanum polysaccharides.
