2014-01-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/709909摘要:基於目前台灣近60 個線蟲相關實驗室對台灣線蟲核心實驗室(CECF)的高度依存與需求,我們在此計劃書中擬規劃延伸與擴充此核心實驗室之服務能量。CECF 於2007年受國科會支持成立以來,在推廣線蟲科學研究已領先全球。我們在促進台灣學界以線蟲作為模式生物進行高端科學研究,已投入許多努力。台灣線蟲社群從初始年產量十數 篇到現今年產量已發表超過30 篇線蟲相關研究,名列全球22,突顯線蟲相關研究在台灣正持續且快速增加。台灣線蟲核心實驗室在2012 年的使用評價調查中使用者滿意評分高達9.35 分。此外,以「線蟲核心設施(C. elegans core facility)」進行關鍵字搜尋,台灣線蟲核心實驗室在全球也名列第一,反映出科學家對本設施的高度需求。本設施目前提供線蟲株保存與分送、各式線蟲表現質體、自動培養皿傾注系統、線蟲株基因型及表型檢測、基因槍使用與全基因體RNA 干擾庫篩選等服務。未來更擬設立CRISPR 基因剔除與線上學習系統之服務。我們未來將開設線蟲生物學相關課程,並舉辦國際級線蟲研討會。我們也已設立線蟲突變或基因轉殖株資料庫、CECF 網站及臉書(Facebook)討論平台,提供研究人員進行資訊交流。為給持續擴大的線蟲研究社群更優質的服, 我們需維持並提升設備與服務品質。因此我們計劃加強現有設施及服務平台、持續提供學術活動,並促進台灣研究人員與國際著名科學家交流。甚者,我們正致力於使台灣線蟲核心實驗室獨立運作,並計畫從2014 年開始收費。因此現階段若能獲得國科會繼續支持,將會是此目標達成的關鍵。<br> Abstract: In this proposal we request an extension of the support for the C. elegans Core Facility(CECF) Taiwan based on its high demand by nearly 60 PIs using C. elegans now in Taiwan.Since its foundation in 2007 (with only 6 PIs using C. elegans in Taiwan) the CECF hasreceived continuous support by the NSC leading to a world-ranked core facility to promote C.elegans research. With enormous efforts by the CECF to facilitate state-of-the-art researchusing this model organism, a visible increase in C. elegans publications can be seen from one dozen in 2007 to more than 30 in 2012. Taiwan is now ranked at position 22 world-wide in publishing C. elegans-based research just after Austria and Denmark. The popularity of C.elegans (a 1 mm long, light-thru nematode with fully sequenced genome and a 3-day life cycle) has indeed increased significantly, especially after the awards of total 3 Nobel Prizes(based on C. elegans research) within 2002 and 2008. During a consumer satisfactory evaluation in 2012 the CECF has received a score of 9.35 (scale: 1-10; 10 = best) from 34 users willing to fill in our evaluation forms. Typing “C. elegans core facility” into the“Google search machine” will immediately lead to the CECF as a first hit, leaving other world-wide worm cores behind, reflecting the high demand and access of our core. Our services encompass bioconservation of animals in large and secure freezers, storage and distribution of Andrew Fire Vectors/RNAi/Fosmids clones, automated plate pouring system, geno- and phenotyping service, RT-PCR and qPCR service, genome-wide RNAi library screening, service for multimode detector microplate reader (for high-throughput RNAi screening), gene gun, organizing C. elegans courses and workshops for students as well as organizing national and international meetings (for example the 5th East Asian Worm Meeting in Taipei in 2012). Recent new services are CRISPR gene knockout service and the establishment of an online learning system (C. elegans E-learning). Importantly, our bioconservation facility holds now 575 C. elegans mutants and transgenic animals that are all organized in a Taiwan-wide accessible database. Besides this database, we also maintain the CECF website (with our service overview and meeting announcements) and a Facebook platform to which (up to now) 84 PIs and students have subscribed facilitating worm and plasmid requests as well as fast exchange of protocols and experimental trouble shooting. Though our past efforts have led to a vast increase of C. elegans researchers in Taiwan, there are still many labs with little experience in worm handling. To provide better services to the exponential growing worm community, we need to continue and sustain the current well running and indispensable services. To strengthen and transform our existing facilities and service platforms and to continuously offer academic activities for promoting interactions among local researchers and international eminent scientists, further NSC support is critical.
