2014-08-012024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/662989摘要:植物交互保護 (cross-protection) 是藉由植物預先感染與強系病毒親緣相近的弱系病毒,進而導致植物產生抵抗強系病毒的抗性反應。在先期國科會計畫中,我們已經建立蕪菁嵌紋病毒 (Turnip mosaic virus, TuMV) 的弱系病毒株 - Tu-GK 與阿拉伯芥 (Arabidopsis thaliana) 為主體的交互保護研究平台。先前的結果顯示,植物交互保護是一種多元性抗病反應 (multiple immunities),其中包含了後轉錄基因靜默 (post-transcriptional gene silencing, PTGS) 作用、蛋白誘導的抗病機制 (protein-mediated resistance) 及植物內生抗性 (innate immunity) 之保護作用。本研究將探討交互保護作用機制中多元抗病性之反應路徑,其主要內容有以下四點:(1) 評估 Tu-GK 誘導之植物內生抗性對不同種及不同屬病毒的抗病能力;(2) 搜集不同抗病路徑的阿拉伯芥轉基因株與突變株;(3) 評估 Tu-GK 誘導之植物內生性抗病反應,在不同抗病路徑的阿拉伯芥轉基因株與突變株中之保護能力;(4) 利用高通量定序分析 TuMV 所感染之獨立免疫反應植株,用以探討不同抗病反應之能力。另一方面,則以病毒累積時程 (time-course pattern) 及高通量定序分析其轉錄體 (transcriptomes) 、病毒小分子核酸 (virus-directed short interfering RNA, vsiRNA) 及降解體 (degradome) 變化,用以探討寄主與病毒間的交互作用,進而深入了解由交互保護作用所引起的多元性抗病反應。<br> Abstract: We developed a Turnip mosaic virus (TuMV) mild strain (Tu-GK) mediated cross-protection system in Arabidopsis thaliana plant in the previous project of National Science Council (NSC). Our previous study indicated that the multiple immunities that include a post-transcriptional gene silencing (PTGS) pathway, protein-mediated resistance, and host innate immunities confer crucial roles in Tu-GK-mediated cross-protection. In this proposal, we plan to further explore the multiple immunity pathways in cross-protection. Four specific goals will be highlighted in this study. (1) The evaluation efficiency of Tu-GK-mediated cross-protection between different species or genera of viruses. (2) Collection of mutants or generation of transgenic Arabidopsis materials in various immunity pathways. (3) An evaluation of the efficiency of Tu-GK mediated cross-protection in various mutants and transgenic plants. (4) The collection of high-throughput deep sequence profiles from TuMV infected samples. The primary purpose is to individually study virus resistance efficiency in various immunities.
