2016-01-012024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/663582摘要:口蹄疫疫苗免疫是控制疾病免於爆發的好方法,但必須監控免疫動物的血清以評估其成效。免疫動物的保護性是與其血清中和抗體力價呈正相關的,因此目前廣為世界所接受可用於檢測口蹄疫免疫動物血清力價的方法便是血清中和抗體力價測定以及液相酵素連結免疫吸附試驗兩種。血清中和抗體力價雖然敏感且準確,但它必須使用活病毒來操作,因此僅能於生物安全第三等級的實驗室內進行。但液相酵素連結免疫吸附試驗卻擁有高敏感性、穩定性以及可同時處理大量樣品的優點,且它的敏感性也與血清中和抗體試驗相同。本計畫擬開發液相酵素連結免疫吸附試驗來取代目前國內長期使用的血清中和抗體試驗來檢測田間血清樣品。<br> Abstract: FMD vaccination is good way to control the disease outbreak, but the immunity level of the vaccinated animal should be monitored. The protection of immunized animal was related with the neutralizing antibody levels of serum. Now the internationally accepted tests for determining the FMD antibody titer of vaccinated animal are the virus neutralization test (VNT) and liquid-phase blocking ELISA (LPBE). The VNT is considered to be sensitive, specific and reliable, but, it must use alive FMDV to takes test. The VNT was restricting in BSL-3 laboratory. LPBE methods have many advantages including high sensitivity, suitability for large scale samples, and the sensitivity is equal to VNT. The project is want to develop LPEB to replace the conventional VNT in Taiwan.
