We aimed to study the in vitro and in vivo efficacy of biodegradable PU nanoparticles loaded with fenofibrate (FNB-PU) on nonalcoholic fatty liver disease (NAFLD). FNB-PU was prepared by a green process, and its preventive effects on NAFLD were investigated on HepG2 cells and mice. FNB-PU showed sustained in vitro FNB release profile. Compared to FNB crude drug, FNB-PU significantly decreased triglyceride content in HepG2 cells incubated with oleic acid and in livers of mice with NAFLD induced by a methionine choline deficient diet, and increased plasma FNB concentration of the mice. FNB-PU increased absorption of FNB and therefore enhanced the inhibitory effects of FNB on NAFLD. ? 2018 American Chemical Society.absorption enhancement; fenofibrate; inhibitory effects; nanoparticles; nonalcoholic fatty liver disease; polyurethane[SDGs]SDG3fenofibrate; nanoparticle; oleic acid; polyurethan; triacylglycerol; antilipemic agent; drug carrier; fenofibrate; methionine; nanoparticle; polyurethan; animal experiment; animal model; animal tissue; Article; controlled study; drug absorption; drug blood level; drug efficacy; drug release; Hep-G2 cell line; human; human cell; in vitro study; in vivo study; male; methionine/choline deficient diet; mouse; nonalcoholic fatty liver; nonhuman; oral absorption; physical chemistry; priority journal; zeta potential; animal; C57BL mouse; chemistry; drug effect; liver; metabolism; nonalcoholic fatty liver; Animals; Drug Carriers; Fenofibrate; Hep G2 Cells; Humans; Hypolipidemic Agents; Liver; Male; Methionine; Mice; Mice, Inbred C57BL; Nanoparticles; Non-alcoholic Fatty Liver Disease; PolyurethanesPolyurethane Nanoparticle-Loaded Fenofibrate Exerts Inhibitory Effects on Nonalcoholic Fatty Liver Disease in Micejournal article10.1021/acs.molpharmaceut.8b005482-s2.0-85053924141https://www.scopus.com/inward/record.uri?eid=2-s2.0-85053924141&doi=10.1021%2facs.molpharmaceut.8b00548&partnerID=40&md5=a4ade282ecfa66967f9c6398b1d3026b
