2015-08-012024-05-13https://scholars.lib.ntu.edu.tw/handle/123456789/649305摘要:1975 年保加利亞,1978 年匈牙利,1997 年馬來西亞,1998 年以來的台灣,以及2007 年後的中國大陸皆有腸病毒71 型的大流行,造成許多人死亡。腸病毒71 型成為致命及重症案例中最重要的腸病毒,尤其是在台灣、中國大陸以及東南亞。另一個重要且有高度傳染力的病毒則是腺病毒,會引起併發症或者是住院治療,甚至在幼童中有重症或是致死的案例。然而目前沒有針對腸病毒71 型和腺病毒感染的抗病毒治療方式。傳統中草藥在這方面有很大的潛力,因為具有多目標的作用機制、較小的副作用以及可能較不會有抗藥性。我們將篩選對抗最重要以及常見的腸病毒(腸病毒71 型、克沙奇A16、以及克沙奇B1 腸病毒)和腺病毒(第1, 2, 3, 4, 7 型)的中草藥。行政院農業委員會臺中區農業改良場陳裕星博士為共同主持人,將提供給我們具有潛力以及穩定來源的中草藥萃取物。我們將篩選多種不同萃取方式的中草藥萃取物,藉由病毒斑(plaque assay)和細胞毒殺試驗(cytotoxicity assay)測定其IC50(50%抑制病毒濃度),以及CC50(50%細胞毒性濃度)以計算出選擇指數(selective index, SI)。根據選擇指數,我們將選出幾個具有潛力的中草藥萃取物做進一步探討其抗病毒作用機制,包括病毒貼附、進入細胞、病毒複製以及釋出病毒能力等。我們希望本研究可以發掘新奇具有抗病毒潛力的藥物,進一步可以幫助預防或是治療重要、具有高致死率並且目前尚未有效抗病毒藥物的腸病毒71 型和腺病毒感染。<br> Abstract: There were large outbreaks of enterovirus 71 (EV71) infections resulting indozens of deaths in Bulgaria in 1975, Hungary in 1978, and Malaysia in 1997, Taiwansince 1998 and mainland China since 2007. EV71 may become the most importantenterovirus related to the fatal and morbid cases, especially in Taiwan, China and thesoutheast Asia. The other important and highly transmissible virus is adenoviruswhich may cause complications or hospitalization, and some severe or fatal cases inyoung children. However, there is no antiviral therapy for EV71 and adenovirusinfection. Traditional Chinese medicine and herbs has shown strong potential becauseit is multi-targeted mechanisms of action, fewer side effects and possible less drugresistance.We will screen Chinese herbs against the most important and commonenteroviruses (EV71, CA16, CB1) and adenoviruses (type 1, 2, 3, 4, 7). The Co-PI, Dr.Chen at Taichung District Agricultural Research and Extension Station, will providethe extracts of potential Chinese herbs because of source stability. We will screenantiviral activities of Chinese herbs including extraction with different methods anddefine the IC50 (50% inhibitory concentration) and CC50 (50% cytotoxicityconcentration) by plaque assay and cytotoxicity assay to calculate selective index (SI)for EV71 and adenovirus. According to SI, we will choose several potentialcandidates to further investigate their antiviral mechanism including attachment, entry,replication, and viral release.
