2012-01-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/659562摘要:本計畫的目標為針對我國最具國際競爭力的番茄、番椒與青花菜等蔬菜作物,開發簡單重複序列(SSR)及單核&#33527;酸多型性(SNP)分子標記系統,配合以具有目標特性之種原所雜交建立的分離族群,建立高密度標記連鎖群圖譜,以定位控制番茄抗晚疫病、番椒雄不稔系統稔性恢復基因、與青花菜早生性之基因座,並將與上述與各作物重要性狀基因緊密連鎖之分子標記,以及具有該基因之衍生育種材料,技術轉移給國內種苗業者或農業研究機構進行分子輔助選種。在前二年的計畫中,已完成(1)產生番椒與番茄F&lt;SUB&gt;2:3&lt;/SUB&gt;進階分離族群及其外表型分析;(2)以自行探勘獲得的SSR分子標記建立番椒連鎖群圖譜並定位稔性恢復基因;(3)以自行探勘獲得的SNP分子標記進行番茄遺傳分析,並定位獲得一組抗晚疫病基因;(4)驗證篩選獲得超過700組青花菜分子標記,其中超過200組在指定親本間可表現多型性;(5)向國內農業研究相關單位推廣以次世代定序系統配合生物資訊學工具開發分子標記。本年度的計畫的主要工作項目包括:(1)栽培青花菜F&lt;SUB&gt;2&lt;/SUB&gt;分離族群,建立完整之青花菜連鎖圖譜,再與調查獲得青花菜早晚生之天數,進行早晚生之數量性狀基因座基因效應分析;(2)找出與&lt;EM&gt;Ph-3&lt;/EM&gt;抗病基因座連鎖更緊密的SNP分子標記,並使用Genotyping-By-Sequencing技術,對番茄F&lt;SUB&gt;2:3&lt;/SUB&gt;進階分離族群進行更多SNP分子標記之基因型鑑定,確認是否存在另外的抗晚疫病的數量性狀基因座;(3)擴大篩選番椒分子標記,增加連鎖群上的SSR分子標記,以期獲得更完整的連鎖群圖譜以及與稔性恢復基因連鎖更緊密的分子標記;(4)舉辦技術發表會向國內種苗業者與農業研究相關機構公布本計畫研究成果,對有意承接之業者或研究機構依技術轉移規範進行技術移轉。<br> Abstract: Objectives of this research are developing simple sequence repeat (SSR) and single sequence repeat (SNP) markers for three most economically important vegetables, namely tomato, pepper and broccoli, in order to construct high density linkage maps to facilitate mapping quantitative trait loci controlling tomato late blight resistance, pepper sterility restoration, and broccoli earliness. Closely linked flanking markers of the target genes as well as the derived lines containing homozygous target genes may be transferred to the vegetable seed industries in Taiwan for markers assisted selection or introgression. Four main tasks in this year are: (1) Constructing broccoli linkage map and mapping quantitative trait loci controlling earliness; (2) Using Genotyping-By-Sequencing method to extend coverage of tomato linkage map, in order to determine the existence of a second late blight resistant gene; (3) Extending the coverage of pepper linkage map and obtaining markers closely linked to the fertility restoring gene, (4).
