2017-01-012024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/664055摘要:隨著飲食和生活型態的改變,全世界肥胖人口逐年攀升,已經成為日益嚴重的健康問題,肥胖同時也會增加罹患許多慢性疾病的風險,例如:第二型糖尿病、高血壓及非酒精性脂肪肝等。目前市面上充斥著許多標榜減肥的商品,雖然可以達到降低體重的效果但通常也伴隨著副作用,因此若能從天然食物中找到具有抗肥胖功效的物質,藉以替代藥物來預防或治療肥胖,將會是保健食品市場的一大商機。紅豆 (Vigna angularis) 富含多酚類、皂鹼及膳食纖維,在先前的研究中指出紅豆萃取物具有保護心血管、預防高血壓及改善胰島素阻抗等功效,因此本研究將使用國產高雄 9 號紅豆,利用四種不同萃取方法,萃取出紅豆之活性成分,並以 3T3-L1 前脂肪細胞評估不同萃取方法之紅豆萃取物對脂肪新生作用的影響, 結果發現,紅豆萃取物無論是常溫水萃取或者加熱水萃取兩組皆可有效減少油滴的形成並使油滴大小明顯變小,其中又以加熱水萃取的組別效果最佳。另外,動物模式以高脂飲食誘導C57BL/6 小鼠肥胖,結果顯示,餵食 1% 及 5% 的紅豆加熱水萃物可以降低小鼠的體重及性腺和腹部脂肪的重量,同時也顯著地降低血液中總膽固醇的含量。在確立紅豆萃取物之抗肥胖功效後,會進行活性指標成分鑑定,並進一步探討紅豆萃取物抗肥胖之機制與腸道菌相的分析。 冀望本研究能證實紅豆之保健功效,且開發成低糖的紅豆益生菌養生片,提升國產紅豆之利用性及經濟價值。<br> Abstract: Obesity problems are associate with the rapid changing social and economic environment. Epidemiological data has demonstrated that high fat diet (HFD) and change in lifestyle linking to obesity, and the prevalence of obesity-derived metabolic syndrome is rapidly increasing in Taiwan and worldwide. Obesity is a chronic condition that caused by imbalance of energy intake and expenditure, results in excessive fat stored in adipose tissue. Excess storage of lipids in adipose tissue is associated with cardiovascular disease, fatty liver and type 2 diabetes. Adipogenesis is a key process in determining the number and size of mature adipocytes in the development of obesity. We investigated the effects of adzuki bean (Vigna angularis) on adipogenesis and the molecular mechanisms in adipocyte differentiation. Furthermore, we examined the anti-obesity and microbiota effects of adzuki bean on obesity animal model. C57BL/6 mice were fed high-fat diet (HFD) and adzuki bean at the same time. The results showed that 1% and 5% adzuki bean extracts significantly reduced mice body weight, perigonadal fat weight and retroperitoneal fat weight in a dose-dependent manner without affecting their food intake. Serum total cholesterol levels and LDL/HDL ratio were significantly lowered for both adzuki bean extracts-treated groups. When the anti-obesity effect of adzuki bean been established, identifying the active compounds from adzuki bean extracts.
