2013-08-012024-05-13https://scholars.lib.ntu.edu.tw/handle/123456789/643243摘要:端粒是染色體最末端的結構,端粒具有保護染色體,維持染色體穩定的作用。端粒上的結合蛋白對於執行端粒的功能是十分重要的。端粒長度的維持對細胞維持染色體的完整性、避免染色體末端之間的連結、以及確保染色體的完整複製是十分重要的。端粒酶是維持端粒長度的的主要蛋白,端粒結合蛋白也參與在調節端粒的長度。除此之外,在酵母菌亦有幾個未知途徑影響端粒長度的維持。本研究主要目的就是要瞭解這幾個未知途徑是以何種機制參與在端粒長度的調節。在過去的研究,本實驗室針對端粒結合蛋白CDC13 的特性作了深入探討,亦發現了幾個可能參與在端粒作用的蛋白。本研究擬持續對端粒長度調節的機制探討,將研究轉錄延長與SUMO 後轉譯修飾對端粒長度的影響。本研究成果將有助於我們對端粒長度的調節有更進一步的瞭解。本研究擬針對以下二方向作深入探討:1、探討轉錄延長在端粒長度調節的機制。2、探討SUMO 後轉譯修飾對端粒重組的影響。<br> Abstract: Telomeres are the structure at the ends of eukaryotic linear chromosomes. They are essentialfor maintaining the integrity of chromosomes. Telomeres protect chromosomes fromdegradation by nucleases, facilitate complete replication of chromosomes, and differentiatelinear chromosome ends from broken ends. Telomere length is mainly maintained bytelomerase. Telomere-associated proteins also participate in telomere length regulation. Inaddition to these proteins that directly affect telomeres, several pathways have been identifiedthat regulate telomere length in yeast Saccharomyces cerevisiae. The goal of the proposedresearch is to elucidate the mechanism of how these pathways regulate yeast telomeres. In thepast thirteen years, we have characterized the telomere binding proteins Cdc13p extensively.We have also identified several proteins that might be components of telosomic complex.Here we propose to continue our efforts to understand how telomere length is regulated. Wewill extend our analysis in two specific pathways including transcription elongation andSUMO modifications. The mechanism of how genes that are involved in transcriptionelongation and SUMO post-translational modification regulate telomere length will beanalyzed. Our analysis should yield valuable information in adding our understanding of themechanism of how telomere length is regulated. Specific experiments are grouped into twocategories:1. To elucidate the mechanism of how THO components regulate telomere function.2.
