2018-01-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/701881摘要:台灣線蟲核心設施(CECF)於2007 年開始獲得補助,在當時全台的線蟲研究計劃與實驗室數目是遠低於全球平均的。在CECF 努力與推廣下,台灣線蟲實驗室數目已增為初期之20 倍。我們開始了CECF 核心入口網站的設立,並樹立了我們的願景目標與服務內容。我們也建構台灣線蟲生物資源保育及生物材料資料庫、鼓勵與推廣學者使用當代線蟲高端的科研方法、並廣邀國外學者進行交流。在國科會(科技部)的補助下,CECF 已購置許多重要的儀器設施與線蟲生物資料庫,包括:自動注膠系統(提供線蟲生物資源之保存)、共軛焦顯微鏡與螢光解剖顯微鏡(提供判斷顯微注射與基因槍製作基因轉殖線蟲株成功產生)、線蟲轉殖載體組、線蟲RNA 干擾基因庫、線蟲全基因體基因庫等各類線蟲生物整合資源,也已供應全台線蟲實驗室所使用。CECF 亦提供基因型及表現型鑑定、RNA 干擾庫篩選與CRISPR 基因剔除等技術平台。單分子螢光原位雜交基因表達分析系統及高解析度溶解分析基因型鑑定系統,也即將上線服務。CECF 除了提供生物資源保存與技術支援外,我們也非常重視學術交流,因此我們也廣邀國外學者來台交流與定期舉辦線蟲討論會與技術工作坊。近期CECF 已邀請德州大學林瑞玲博士(台大14/07/29)與名古屋大學高木新博士(台大14/03/03)進行線蟲教學與技術交流。此外我們也已舉辦過數場國際研討會,其中於2010 年舉行之第一屆台灣線蟲會議與2012 年之第五屆亞太(東亞)線蟲會議最受注目,在本期計劃中,我們也擬議籌備第二屆台灣線蟲國際研討會。為了更有效地傳承線蟲研究之相關知識與技術,在本期計劃中我們也擬建立一數位學習平台,並且將過往與爾後所舉辦之教學與實驗技術工作坊,錄影後置並載入該數位學習平台。在本期計劃中,我們也將探討強化設施及服務收費之可行性,俾使CECF 能獨立運作、永續經營。CECF 已憑藉自動注膠系統獲取部分收入,相信未來其他設施亦能挹注報酬提供營運。以現有設施支援目前台灣的70 間線蟲實驗室,甚至未來將新成立的實驗室,實為CECF 當前最大挑戰。這也彰顯持續強化CECF 服務設施,並得到長期資助,以確保CECF 高品質服務的重要性。<br> Abstract: The first funding for the CECF (C. elegans Core Facility Taiwan) was received in 2007 at a time C. elegans research was conquering labs by storm world-wide while being extremely underdeveloped in Taiwan. Based on the enormous efforts of the CECF to make available this important type of model organism to labs in Taiwan until now the number of worm-labs has been increased by a factor of twenty. We started with setting up a website announcing our goals and missions and offering our services. Following a database made ac-cessible to all Taiwanese labs using C. elegans for bio-conservation, archiving and distribution strains and plasmids. We actively hired PIs using state-of-the- art methods and invited our collaborators from abroad to exchange and transfer expertise. We received support for the purchase of important equipment as automated plate pouring system (for worm maintenance and conservation), as well as confocal microscopes and stereo fluorescence microscopes to monitor successful gene transfer in our microinjection and gene gun facilities. Financial support was also given to purchase important libraries as the “Andrew Fire vector collections”, “RNAi feeding clones” and the “Fosmid library” all made available to the worm community. We then estab-lished additional services such as geno- and phenotyping services, genome-wide RNAi library screening ser-vice and CRISPR service. In this proposal we describe our new service “single molecule fluorescence in situ hybridization (smFISH) in C. elegans” which we are going to offer to the community. Besides providing these important services, we must transfer knowledge and expertise to PIs and students to fulfill our goals and missions (to promote C. elegans research in Taiwan). Thus, we frequently organize seminars and work-shops and invite international speakers. Just recently, we invited Dr. Rueyling Lin (from UT Southwestern) who held a whole day workshop on “C. elegans Genetics” (July 29th 2014 at NTU) and we also invited Dr. Shin Takagi (Nagoya University) to give a special seminar on novel “Optogenetic tools” (March 3rd 2014 at NTU). The list of held seminars and workshops is long and notable are the “1st Taiwan-wide C. elegans meeting” in 2010 as well as the “5th Asian-wide C. elegans meeting” in 2012. In the course of this proposal, we will organize the second Taiwan-wide C. elegans meeting. To educate our students, we are in the process of establishing an E-learning platform and video-taped workshops are currently in the post-production pro-cess. We will accomplish our task to provide web-based seminars for free to all students in Taiwan easily ac-cessible from everywhere in the course of this proposal. We also discuss strategies how to strengthen our current services and to prepare them for charging fees inevitable to become a self-sustained core facility. We made our first revenue with our automated plate pouring system and we are confident that users are willing to reimburse for other services likewise. Taiwan now holds 70 labs using C. elegans as a model organism and the CECF is facing a vast challenge to support these labs with our services. Moreover, there are still many labs with little experience in worm handling.
