2003-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/705556摘要:紅麴是傳統上廣泛使用之天然食用色素,近年來由於發現具有多種保健功能而受到重視。已經證實之有益人體之成分包括膽固醇合成抑制劑monacolin K、降血壓成分γ-aminobutyric acid (GABA)及抗氧化物質dimerumic acid等。然而,紅麴代謝物中亦含具腎毒性之citrinin。紅麴製品雖已普遍地商品化販售,但是其化學成分極為複雜,且仍有許多未知之生物活性仍需進一步地研究與探討。我們初步的研究結果發現,紅麴丙酮萃取物之特定區分物(橘色系azaphilone compounds)對於肺上皮癌細胞株A549具有顯著促進其增生之生物活性,但對正常細胞---人類胚胎肺纖維母細胞株MRC-5之增生卻無影響。這顯示此成分有可能在上皮細胞(epithelial origin)癌化的過程中,促進上皮細胞增生或延長上皮細胞存活,因而促進腫瘤(carcinoma)之形成。本研究擬針對紅麴代謝物中具有此生物活性的區分提出二年期的研究計畫,探討的項目包括:對此生物活性成分進行分離與純化,並進行相關促進生長之活性評估。<br> Abstract: Anka is prepared by culturing Monascus sp. on rice grains. Traditionally, anka pigment is used as natural food colorant. Recent discoveries indicate Monascus products with multiple health effects and determine many biofunctional compounds, such as cholesterol synthesis inhibitor, monacolin K; hypertension controlling substance, γ-aminobutyric acid (GABA); and antioxidant, dimerumic acid. However, the nephrotoxic citrinin also exist within the Monascus products. Although anka is already marketed as health supplement, its complexity in contents remains numerous uncertain biological activities deserved for further research. According to our preliminary research work, the fraction of anka acetone extract performed the mitogenic effects to the lung carcinoma A549, but no significant effects to the normal fibroblast MRC-5. It is suggested that during the process of tumorigenesis of epithelial cell, the mitogenic factor might promote proliferation or extend the survival of epithelial cell. Furthermore, it might also increase the potential for carcinoma formation. Based on the above, the present project will be carried out in two years to clarify the mitogenic effects of anka extract. The subjects are respectively as follows: (1)Separation and purification the fraction of the mitogenic factor from anka extract.
