2011-01-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/682397摘要:Potyviridae科Potyvirus屬病毒為長絲狀、單鏈正股RNA病毒,是植物病毒中最大的一屬,在田間由蚜蟲以非永續方式傳播,對農業造成相當大的損失。若能發展出一套可快速且成功檢測及鑑定各種potyvirus的方法,對於各國的農業以及植物防檢疫工作應有極大的助益。本計畫希望研發出可快速檢測potyvirus之ELISA和快檢試紙套組,以及蕪莖嵌紋病毒(TuMV)和海芋微嵌紋病毒(ZaMMV)兩種potyvirus鑑定套組,以協助檢測植物樣品中是否含有potyvirus,並可鑑定是否有TuMV或ZaMMV存在。本計畫今年將執行下列工作項目:(1) 大量製備和純化TuMV和ZaMMV之專一性多株抗體,(2) 決定TuMV和ZaMMV的ELISA檢測套組之產品型式,(3) potyvirus快檢試紙之改良與調整,(4) ELISA檢測套組的實際測試。所研發之potyvirus快速檢測套組包括ELISA和快檢試紙產品,可提供國內研究與防檢疫單位測試使用。<br> Abstract: The genus Potyvirus (in the family Potyviridae) contains the largest number of plant virus species which have single-stranded, positive-sense RNA genomes with filamentous particles. Potyviruses are transmitted by aphids in a non-persistent manner and cause significant losses in a wide range of crop plants. If the rapid detection and identification kits are developed for potyviruses, the agricultural industry and the works of plant inspection and quarantine will greatly benefit from them. The goals of this project are to develop the rapid detection kits including potyvirus immunostrip and ELISA kits for Turnip mosaic virus (TuMV), Zantedeschia mild mosaic virus (ZaMMV) and potyviruses. By using these kits, potyvirus-infecting plant samples can be detected and the presence of TuMV and ZaMMV can also be confirmed. In this year the following works will be executed: (1) Large amount of polyclonal antibodies against TuMV and ZaMMV will be separately prepared and purified. (2) The format of ELISA kits for TuMV and ZaMMV will be determined. (3) Potyvirus immunostrip will be modified and improved. (4) All of the ELISA kits will be tested.
