2017-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/706419摘要:臺灣紅藜是臺灣本土的傳統植物,紅藜的營養價值與國外的藜麥相似,皆屬於高蛋白且完整胺基酸組成之作物,而其顏色則與莧菜籽相仿,都含有紅色的甜菜色素。近年來在地食材受到重視,紅藜已廣泛應用於飲料、麵包中,然而,對於紅藜的化學成分報告較少,而同為藜麥屬的藜麥、莧菜籽因外觀與紅藜相仿,容易混用而誤導消費者。因此,本研究將從化學成分的角度,試圖找出臺灣紅藜的身分鑑定指標,例如特殊的酚酸、類黃酮、皂素或固醇的組成。本計畫將利用高解析質譜儀之代謝質體學的研究模式,先採用特定目標物的分析方式,探索臺灣紅藜中酚酸、類黃酮、甜菜色素、皂素及固醇類的組成,再以非特定目標的分析方式搭配多變量統計方析,建立臺灣紅藜的指紋圖譜,作為身分鑑定的標準。為了全面了解臺灣紅藜的化學組成,將針對不同極性的化合物分年研究,規劃第一年針對中高極性的物質進行探討,研究紅藜中酚酸、類黃酮的組成與含量;第二年針對高極性物質,包含甜菜色素及皂素,進行定性與定量分析;第三年針對低極性的植物固醇及脂溶性維生素進行方法開法;此外,每個極性區分物都會進行指紋圖譜的分析。希望能藉由代謝質體學的研究方式,建立鑑別臺灣紅藜、藜麥及莧菜籽三者之化學分析方法,作為臺灣本土紅藜之身分鑑定標準。<br> Abstract: Djulis (Chenopodium formosanum) is a traditional cereal plant as food source used by Taiwan indigenous people. Djulis, similar to the quinoa (Chenopodium quinoa), is a highly nutritious food with the high content of protein, and its color comes from betalains so does amaranth (Amaranthus hypochondriacus). Locavore (locally grown ingredients) is taken more attention, recently. Djulis has widely used in food, including beverage and bread. However, there are limited researches on the chemical profile of Djulis. In addition, the physical appearance of quinoa and amaranth were similar to Djulus, thereby the consumers may confuse because of adulteration. The aim of these projects is to characterize the authentic of Djulis via the chemical composition. High-resolution mass spectrum-based metabolomics analysis was carried out on food authenticity. The first step is targeted analysis by evaluating the chemical profiling including phenolic, flavonoids, betalains, saponins and phytosterols in Djulis. The second step is non-targeted analysis by characterizing the fingerprinting of Djulis by chemometric analysis. To comprehensive identify the chemical composition of Djulis, the different polar molecules were divided and evaluated in three years. The first year focus on the medium-high polar molecules, including phenolic and flavonoids. The second year focus on the high polar molecules, including betalains and saponins. The third year focus on the lower polar molecules, including phytosterol and fat-soluble vitamins. Moreover, the fingerprinting of each polarity fraction was performed.
