2014-05-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/657099摘要:在爆發傳染病流行後,隔離檢疫自然是有效的防治之道,然而現代社會人與人接觸頻繁,每日往返世界各國間的商務或旅遊人士日以遽增,造成消極的隔離做法難以完全阻絕傳染病的橫行,而重大傳染病的流行除造成生命的威脅外,亦可能引發國家重大損失。有效的疫苗是目前公認預防傳染性疾病最有效的方法,所以對於疫苗之研發與製造必須從國家安全的層級考量,亦應有長遠計畫,規劃疫苗產業發展方向,進而確保國家安全。 分子生物學及免疫學快速發展,應用不同於傳統疫苗產製方法所製造的新型疫苗已陸續被開發出來,更成為新興的經濟產業。我國在1) 於感染症的臨床和基因研究上,已有深厚的基礎;2)常見癌症與感染症的特質,與世界已開發國家不同及3) 建立長期觀察研究的跨領域團隊之優勢,藉由國家型計畫與任務型優先推動中程計畫長期的推動與研發下,已累積了厚實的能量與基礎且持續推動中。 本計畫以達成「落實保健防疫整備,免除疾病威脅」與「發展醫藥衛生科技」之策略目標為起點,開發前瞻疫苗技術,新型疫苗及免疫治療藥物,發展我國疫苗自製能力,執行疫苗相關之國家政策,因應新興傳染疾病疫苗之緊急需求,以提昇台灣甚至亞洲地區之疾病預防與健康照護水準,並藉由整合國內有限資源與研究人才,建立國家級疫苗研發團隊,投入重要疫苗技術之研發與量產,本計畫辦公室運作計畫延續計畫擬依據原擬定之發展方向包括:1.建立可產生廣泛及高抗體中和反應之技術 (Technology can generate broad and high affinity neutralizing antibody responses)。2.新型佐劑研發(Novel adjuvant discovery)。3.發展可提昇細胞培養病毒或重組蛋白產能之生物製程(Bioprocess can enhance cell-based virus or recombinant protein production)。4.發展反轉錄疫苗學、反轉錄基因學與微生物合成技術(Reverse Vaccinology, Reverse Genetics and Synthetic Microbes)。5.發展可提升黏膜細胞免疫之載體技術(Vector technology for mucosal immunity)。6.新疫苗發展的倫理、法律及社會的影響之議題研究。以強化疫苗研發基礎,建立我國自製疫苗之能力,因應本土特殊傳染病應變與防疫能力。更進一步,鼓勵疫苗產業發展,降低本國依賴國外進口的情形,使國內疫苗製劑安定供應,另更期望能於未來發展「疫苗外交」推展外交實務,厚實我國之國際地位。 <br> Abstract: After the outbreak of infectious disease, quarantine is effective control of the epidemic disease. However, frequent physical contacts between people and international travel, nowadays, caused difficult to completely quarantine people from epidemic diseases. When emerging infection diseases spreading, it not only threatens the peoples’ life, but also may cause huge national damage. Vaccine is widely known as the effective prevention of infectious diseases. Thus, the development and manufacturing for the vaccine is important to ensuring national security. Because of rapid development on molecular biology and immunology, new vaccines have been developed apart from the traditional manufacturing method. In Taiwan, the study in the infectious disease of the clinical and genetic research has a good foundation to develop the new vaccine. This continued program project is to keep supporting the research and development proposals in following topics: 1) Technology can generate broad and high affinity neutralizing antibody responses. 2) Novel adjuvant discovery. 3) Bioprocess can enhance cell-based virus or recombinant protein production. 4) Reverse vaccinology, reverse genetics and synthetic microbes. 5) Vector technology for mucosal immunity. 6) The ethical study of new vaccine development and legal and social implications.
