2018-05-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/692121摘要:新型豬流行性下痢病毒(Porcine epidemic diarrhea virus; PEDV)自 2013 年於北美及包括台灣之亞洲 各國爆發以來,疫情尚無法有效的控制,重創各國畜產業發展,也是造成台灣豬價上揚的主因,嚴重 影響民生經濟。因此,豬流行性下痢病毒之疫苗開發為目前當務之急。有鑑於這幾年在亞洲所爆發的 豬流行性下痢疫情,推測有可能是使用未減毒完全或失活之 PEDV 所造成的,因此 PEDV 次單位型疫 苗的研發是相當重要且急迫的。為能有效地於短時間內開發出可商業化之 PEDV 次單位型疫苗,本計 畫整合兩項新穎的桿狀病毒表現系統:(I)以多效桿狀病毒表現系統於昆蟲細胞生產 PEDV 病毒的類 病毒顆粒(virus like particle, PEDV-VLP)疫苗(子計畫一)及(II)以桿狀病毒生產並展示豬流行性下痢病毒 表面抗原棘蛋白(spike protein)之重組桿狀病毒,稱為 S-Bac,以為新型疫苗 (子計畫二),接著由在 PEDV 動物實驗已有成熟經驗的子計畫三建立 PEDV 疫苗動物實驗測試模式(vaccine animal model),以試驗出 最有效的疫苗。本計畫將發展可商業化的安全且經濟的 PEDV 病毒疫苗,並將進行以此兩不同配方疫 苗誘導田間豬隻免疫及保護力測試實驗,以篩選出最有效的疫苗配方,並應用於母豬以誘導其產生移 行抗體及被動免疫保護新生小豬免於 PEDV 致害,提高仔豬育成率,促進國家畜產業及民生經濟恢復。 同時我們也將開發出新式簡易高效的中和性抗體檢驗分析方法以有助於分析疫苗效力,將來也能分析 豬隻感染的疫情。更重要的是本計劃所建立之新穎高產量且安全之疫苗產製方法為一平台式科技 (platform technology),除可快速應付新突變爆發的 PEDV 疫情外亦可應用於其他病毒性疾病之疫苗發 展。本計畫也將對畜牧產業受到不斷感染的環境逆境提出一個重要的解方。<br> Abstract: Since 2013, new porcine epidemic diarrhea viruses (PEDVs) have rapidly spread to North America and Asia, including Taiwan. The disease severely impacts swine industry in these countries resulting in severe economic loses. Up-to-date, there is no effective PEDV vaccine in controlling the disease. A new generation of PEDV vaccine is therefore urgently needed. For a novel, effective, and economic PEDV vaccine development, we aim to develop subunit vaccines using two novel strategies in production of immunogens with high quality and quantity. We will generate virus like particles, PEDV-VLP (sub-project I) as an effective vaccine, we will also design and use baculovirus to display the spike protein of PEDV on the envelope of baculovirus (named S-Bac, sub-project II) as a novel effective and easy to manipulate vaccine. The immunogenicity and protection efficacy of these two vaccine candidates carrying glycosylated proteins by the insect cells expression system against PEDV will be evaluated in conventional postweaning pigs (sub-project III). The best vaccine formulation will be selected and applied in pregnant sows to induce systemic and passive immunities to protect nursing piglets. We will also develop novel assay system to analyze neutralizing antibodies to study the efficacy of different vaccines. The project will provide effective and safe vaccines in protection of pigs from PEDV infection and improve the development of swine industry and economy. The novel techniques will also serve as good tools for other viral (coronaviruses) vaccines development.
