2015-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/714282摘要:在台灣及世界各國畜養之仔豬目前正遭受嚴重新型豬流行性下痢病毒( Porcine Epidemic Diarrhea Virus; PEDV)感染,本病毒導致不同年齡豬隻嚴重下痢及仔豬高死亡率之疫情,不但造成養豬業者經濟損失,更造成豬價飆漲並衝擊民生。由於目前所知市面上疫苗株(CV777及DR13)無法保護此新興病毒株之感染,新一代的PEDV疫苗之研發及產製有其迫切之需求。由於此新型PEDV台灣病毒株之相關資料有限,本計畫首先將分離田間新型PEDV及定序其S基因,以暸解台灣PEDV新型病毒株與各國分離株之相關性,以提供防治策略方向之擬定,接著將以所分離之台灣PEDV病毒株製成馴化病毒疫苗,進行口服免疫以評估其誘導中和抗體免疫能力與疫苗保護效力,期許能保護豬隻免於目前流行之新型病毒株之感染。同時為了能快速產製並研發具有廣效的保護不同PEDV分離株之疫苗,本計畫亦將同時構建及產製以trimeric S蛋白及其受體結合區(receptor-binding domain; RBD)蛋白之次單位疫苗,期望研發出快速安全之新一代疫苗供國內外田間豬場防疫流行性下痢疾病之使用。<br> Abstract: Pig industry around the world has a longstanding widespread occurrence of porcine epidemic diarrhea virus (PEDV) infection. Recently, a mutated PEDV has led to a serious epidemic in many countries, including in Taiwan and neighboring countries and in the USA. The affected piglets showed severe diarrhea and high mortality leading to a significant economic loss in these countries. It has been demonstrated that currently available vaccines, CV777 and the attenuated PEDV DR13 vaccines are not able to protect against the infection and control disease progression from the new PEDV isolates. A new generation of PEDV vaccine is therefore urgently in need. As very little is known about the new circulating PEDV strains in Taiwan, our initial step is to isolate and to sequence S gene of Taiwanese new PEDV strains. The obtaining result could not only provide relevant and crucial information to understand the epidemiology of emerging PEDV strains in Taiwan, but also be used for identifying potential candidates for developing attenuated PEDV vaccine to prevent the infection caused by current PEDV strains. On top of above-mentioned plans, we also plan to construct and generate the Spike (S) trimeric protein and the receptor binding domain (RBD) subunit vaccines.
