2004-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/705213摘要:由植物萃取具有生物活性及保健醫療等功效之成分,為近年來天然物研究的重要發展方向之一,計劃主持人曾分析台灣五木中台灣杉之抽出成分,發現台灣杉具有各種生物活性及抗癌活性成分,此結果顯示本土植物中的許多天然物具有醫療保健之潛力,值得進一步研究與開發。臺灣肖楠亦為臺灣貴重針葉五木之一,經由初步預備試驗結果得知其萃取物具有良好的生物活性,故本研究計畫之目的為萃取與分離臺灣肖楠抽出成分,評估其各種生物活性,包括抗真菌、抗白蟻、抗細菌、抗室塵、抗病媒蚊、抗氧化、抗腫瘤等,並鑑定具生物活性成分之化學結構。首先,以溶劑萃取臺灣肖楠各部位之抽出物,續以不同極性之溶劑萃取區分為各分離部,並進行各項生物活性試驗,篩選出各種生物活性效果最佳之分離部,再經由層析技術加以分離與純化,所得之純化合物則利用核磁共振光譜、質譜及紅外線光譜等儀器分析加以鑑定化學結構,同時亦將純化合物進行各項試驗以便確認其生物活性;另外,亦將進一步解析純化合物對不同生物或腫瘤細胞之抑制機制。本計畫如能順利完成,將可了解臺灣肖楠萃取物的各項生物活性,並使具有生物活性之成分應用於醫療及保健等用途,進而達到擴展森林產物利用領域及提高經濟價值目的<br> Abstract: Extraction of natural compounds having specific bioactivies and/or medicinal properties from plants is one of the important developments of natural products researches. Recently, the bioactivities of the extractives from Taiwania cryptomerioides, one of the five precious woods in Taiwan, were evaluated and some potential antitumor compounds were identified in our laboratory. It reveals that many natural products existed in the endemic plants are of great potential as bioactive agents or pharmaceuticals. Further researches and developments in natural products are worthy to exploration. Calocedrus formosana Florin is also one of the five precious woods in Taiwan. According to our preliminary study, the bioactivity of C. formosana extracts is commendable. Therefore, targets of this project are to evaluate the bioactivities of C. formosana extracts and to isolate the bioactive pure compounds. At the beginning, different parts of C. formosana will be extracted with various organic solvents. The extracts are then separated into several subfractions by liquid-liquid partitions. To obtain the best active subfractions, every subfractions collected are under the examination for various kinds of bioactivities, including antifungal, antitermite, antibacterial, antimite, mosquito larvicidal, antioxdiant, and antitumor activities, etc. The pure compounds existed in the subfractions are finally isolated by chromatographic separation techniques and identified by modern spectroscopic analyses, such as NMR, MS, FTIR, etc. Identified compounds are also examined with bioactive assays to confirm their bioactivities.
