2019-08-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/677437摘要:生物3D列印是近年來興起的一種技術,其結合組織工程與3D列印技術製作各種結構與形狀的生 物支架,可應用於修復或重建人體組織和器官。然而目前的生物3D列印機台與材料都各有其缺點,且細胞在列印的過程中亦可能因剪切應力過大導致細胞受損或死亡,因此細胞列印材料與機台的配合為積層製造生物3D列印的關鍵技術。本計畫有五個主要標的:(1)有別於前一期所開發之生醫材料低溫3D列印機台,本期計畫將建立可變多噴頭於常溫與體溫下列印不同材料與細胞組合之系統。(2)開發3D深度相機軟體技術,擬重建出傷口處原貌,並經由積層製造系統製造型態吻合傷口之生物支架。(3)開發生物體內可降解之含細胞列印材料,使列印後的細胞在列印材料中仍可保持活性。(4)透過糖尿病大鼠模型模擬皮膚受損後,藉由3D列印人工皮膚之治療方式證實優於傳統治療方法。(5)結合細胞、材料與3D列印技術,以人工皮膚為載具開發具產業化潛力之製程。透過本計畫之執行,期望能利用所開發之生物列印材料、積層細胞列印系統與3D深度影像重建之技術,進行人工皮膚與其他組織之客製化生物支架列印,並將整套系統以技術授權或新創公司之形式產業化。<br> Abstract: Three-dimensional (3D) bioprinting is an emerging technique to create tissue-like structures for tissue or organ repair and reconstruction. However, the printing system and materials (bioink) commonly utilized for 3D bioprinting have their drawbacks. In addition, the shear stress during the process of printing cell-laden constructs may induce cell damage or cell death. Therefore, the combined printer and material design plays an important role in additive manufacturing for tissue engineering. In this project, our specific aims are: (1) different from the low-temperature platform developed in the previous years, we will establish the 3D printing platform and system for bioprinting at room temperature or body temperature; (2) we will develop the 3D digital photography system for rebuilding the morphology of the injured area and further print the same size of constructs by the bioprinter; (3) we will develop a series of bioinks and combine with the bioprinter for printing cell-laden constructs. The printed tissue constructs will be further applied as skin substitutes; (4) there skin substitutes will be employed to treat skin injuries of diabetic rats and compared with the traditional treatments; (5) we will develop an industrial process for manufacturing artificial tissue.
