2010-01-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/659322摘要:本計畫利用近年發展之基因體技術研究早期胚基因表現差異,探討胚細胞基因表現之調控機制,獲得之成果則有利於開發母豬繁殖性能的免疫呈色檢測套組及具差異表現之基因型檢測套組,冀能實際與快速地縮短種豬選育時程,並藉此提升豬隻繁殖效能。預期將已產製之目標基因重組蛋白質抗原依胚期及生物功能重新分組為2至3群以適應設計之檢驗套組型態,並進行以酵素免疫呈色方式進行與抗體結合效率、濃度決定等之先導實驗,根據先導實驗結果補充並產製所需之相關目標基因與internal control genes之蛋白質抗原與抗體。在種豬場收集種母豬繁殖性能與品系整體資料,選出高繁與低繁性能母豬群,進行發情同期化並取胚,取得樣品後製備,進行晶片檢測其基因表現差異,並予以分析獲取高繁與低繁性能表現相關的差異表現基因群。本計畫預計完成一組母豬繁殖性能蛋白質測試檢驗套組一組與差異基因regulatory SNP檢測套組一組,以便針對不同繁殖性狀之母豬進行測試,另外也必須完成基因轉錄/轉譯、細胞分化/發育、基礎功能與能量傳遞相關等的表現差異基因分群,提供更豐富的母豬繁殖性能資料。<br> Abstract: In this project, the genomic technology for detecting differential gene expression at the early stages of porcine embryogenesis was used to study the regulatory machnism of the gene expression in embryo. The results obtained will profit the development of ELISA-like test kits and genotyping array of differential gene expression for examining sow reproductive performance. The testing kits are expected to improve the sow reproductive efficiency and to shorten the selection procedure of breeding pigs. Those recombinant protein antigens of the target genes will be grouped to 2 to 3 groups to accomdate the design of testing kits. The pilot experiments of ELISA will be conducted to test the binding efficiency of antibody and concentration to produce and supplement the antigen and antibody. The performance data of sow reproduction will be collected from the cooperated breeding farm to select sows of high and low reproductive performance to feed different diets and mycotoxin. Those sows will be flushed to collect embryos, and then prepared to conduct the gene expression experiments using microarry to find out the differential expression genes. The project for this year is expecting to accomplish a preliminary protein testing kit for sow reproductive performance to practically test in the field.
