國立臺灣大學植物病理與微生物學系暨研究所劉瑞芬2006-07-262018-06-292006-07-262018-06-292004http://ntur.lib.ntu.edu.tw//handle/246246/17693疫病菌(Phytophthora parasitica Dastur, Phytophthora nicotianae Breda de Haan)不僅寄主 範圍廣泛,造成的危害也極為嚴重,是非常重要的植物病原真菌。近年來關於疫病菌誘導 性物質及其在植物可能引發之抗病反應的研究頗多,但對於疫病菌因應植物訊息的傳入會 啟動哪一類訊息傳導途徑,並引發哪些致病相關基因的表現目前仍不清楚。為了探討訊息 傳導途徑mitogen-activated protein kinase (MAP kinase) pathway 在疫病菌致病過程之重要 性,本研究首先進行疫病菌MAP kinase 基因之選殖與特性分析。研究結果顯示,疫病菌至 少含4 個MAP kinase 基因,分別命名為ppmk1.1, ppmk1.2, ppmk2 與ppmk3,其胺基酸序 列與其他病原真菌之MAP kinase 具有相當顯著之保守性。基因組南方雜合分析之結果顯 示,疫病菌各含單一copy 之ppmk1.1, ppmk1.2 及ppmk3 基因,但除ppmk2 基因外,可能 還有其他ppmk2 同源性序列。這些MAP kinase 在疫病菌生理、生活史及致病過程可能扮演 之角色仍待進一步探討。Phytophthora parasitica Dastur (=Phytophthora nicotianae Breda de Haan), an oomycete plant pathogen, caused severe diseases in a wide variety of plant species. In eukaryotes, the MAP kinase pathways are activated in response to a wide variety of extracellular signals, and thereby regulate the processes of growth, differentiation, and pathogenesis in many organisms. As a first step to investigate the role of MAP kinase pathway in the pathogenesis of P. parasitica, full-length cDNA encoding MAP kinase was obtained by reverse transcription-PCR and rapid amplification of cDNA ends (RACE). A total of 4 genes were identified, designated as ppmk1.1, ppmk1.2, ppmk2, and ppmk3, respectively. Analysis by genomic Southern hybridization revealed the existence of a single copy of ppmk1.1, ppmk1.2, and ppmk3, respectively, in the genome of P. parasitica. However, more than one band were detected with ppmk2, suggesting the presence of other ppmk2-like sequence. Analysis of the deduced amino acid sequences indicated that the MAP kinases of P. parasitica were highly conserved with those of fungal pathogens.
