2009-09-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/660509摘要:人類與大部分的模式生物已進入後基因體時代,但仍有許多研究上重要物種仍處於有限的基因體序列與基因功能資訊未完整解碼狀態,這是本校相關研究團隊之研發可以領先國際的地方。鑒於生農學院、生命科學院與其他學院四十多個研究室有分析序列或基因型與分子標記的需求,本校「生物技術研究中心」與「系統生物學及生物資訊學研究中心」跨領域合作提出一個二合一的核心實驗室:高通量分子標記檢測與基因/啟動子預測及基因功能註解核心實驗室,希望為全校各研究團隊或實驗室提供整合性的分子標記檢測(以SNP為主其他為輔)及其相關之生物資訊服務。同時希望整合原有的DNA定序儀,加上新購一台高通量分子檢測儀和即時PCR分析儀以服務本校各研究室。也希望藉此建立合作橋樑,增進老師們的研究合作。而利用國際已知的基因體蛋白質體資訊,透過生物資訊演算法預測基因體上的功能區域,提供各物種研究團隊建立基礎資訊,並藉由上述基因體基礎資訊,提供研究團隊進行未知SNP分子標記位置點的驗證序列與PCR引子的設計,當SNP位置點確認後,能夠快速提供研究團隊樣品之基因型的訂定,提供基礎與檢測結果資料整合,促進研究團隊的研究效能。此核心實驗室預計協助不同研究團隊進行五個以上物種之基因體序列分析、新穎SNP位置點驗證與已知SNP位置點基因型的訂定,並建立三個以上這類物種之基因體序列與功能註解資料庫,此外,核心設施亦可提供服務給植物、動物與微生物分子標記研發人員,配合各研究團隊的需求加入分子標記(SNP位置點)資訊,藉此得以直接提供各研究團隊發表文獻之資料圖表所需數據,增進各團隊之研發效能。<br> Abstract: While human and most of those model organisms move into so called “post-genomics era”, many species with certain importance in scientific research are still left in limited information of genomic sequence and/or gene function annotation. The research teams working on those species in NTU should be able to produce important influences in the international scientific platform at this moment. In order to serve the needs from many laboratories in the College of Bioresources and Agriculture and the College of Life Science, the Center for Biotechnology and Center for Systems Biology and Bioinformatics in NTU cooperate to propose a two-in-one core facilities: High-throughput Molecular Marker Core and Gene/Promoter Prediction and Gene Function Annotation Core. These two core facilities are able to provide those research teams integrated services for sequencing and molecular marker analysis (mainly SNP site validation) and genotyping as well as their related bioinformatics applications: utilizing the international public databases of genomic and proteomic information to predict the functional sites in the genomic DNA sequences using bioinformatics tools; providing the sequences around certain functional sites to validate novel SNP sites with the basic information in the previous step; genotyping those validated SNP sites using high-throughput SNP machine. Those results will be integrated into a database together based on the design from research teams for publication and further studies. There will be analysis of sequences, novel SNP sites validation, and genotyping in more than five non-human and non-model organisms.
