2006-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/710286摘要:國人的肝臟疾病罹患率甚高,許多的肝臟疾病(如慢性肝病、肝硬化及肝癌)名列於國人十大死亡原因之中。且肝臟在人體中扮演著執行代謝、解毒與抗氧化的角色,因此如何保持肝臟的健康在台灣是一個重要的課題。此外,目前已有許多研究證實真菌類(如靈芝和巴西洋菇)與豆科植物(如黑豆和黃耆)中,存有多種機能性成分,能調節人體生理活性。在我們之前的研究成果指出,在離體實驗中「靈芝-豆科」或「巴西洋菇-豆科」發酵菌絲體之乙醇粗萃物皆具有不錯的抑癌效果。 本研究之目的為以動物腫瘤模式探討「靈芝-豆科」或「巴西洋菇-豆科」發酵菌絲體乙醇粗萃物之抑制肝癌活性,並藉由腫瘤中基因的表現與癌細胞的再培養探討樣品抑制肝癌之作用機轉,期能對於「靈芝-豆科」或「巴西洋菇-豆科」發酵產物之機能性能有深入瞭解,以作為健康食品開發之重要參考。本研究擬分兩年進行: 第一年:建立動物腫瘤模式,將人類肝癌細胞株移植到裸鼠身上。探討「靈芝-豆科」發酵菌絲體乙醇粗萃物之抑制肝癌活性。此外,藉由腫瘤中基因的表現與癌細胞的再培養探討樣品抑制肝癌之作用機轉。 第二年:探討「巴西洋菇-豆科」發酵菌絲體乙醇粗萃物之抑制肝癌活性。此外,藉由腫瘤中基因<br> Abstract: The incidence rate of liver diseases is high in Taiwan, such as chronic hepatitis, liver cirrhosis and hepatoma in the top ten causes of death. Liver is the major organ in human being for metabolism, detoxification and antioxidation system. Therefore, how to keep a healthy liver is a big issue here in Taiwan. Recently, many researches have reported that fungi (ex. Ganoderma lucidum and Agaricus blazei) and leguminous plants (ex. Black bean and Astragalus membranaceus) contained many active compounds which can manipulate biological activities. In the previous study, our results indicated that the ethanolic extracts of mycelium of G. lucidum and A. blazei cultivated in the medium containing leguminous plants gave high anti-hepatoma activity in vitro. The objectives of this project are to investigate the anti-hepatoma activity of the ethanolic extract of mycelium of G. lucidum and A. blazei cultivated in the medium containing leguminous plants using tumor-bearing animal model, and to study the mechanism of anti-hepatoma activity by gene expression in tumor and reculture of hepatoma cells from tumor. Hopefully, the physiological functionality on liver protection of the fermentation broths of G. lucidum and A. blazei cultivated in the medium containing leguminous plants will be elucidated and used as important reference for health food development. This project will be performed in two years: First year: To establish a tumor-bearing animal model, nude mice will be inoculated with human hepatoma cells. To investigate the anti-hepatoma activity of the ethanolic extract of mycelium of G. lucidum cultivated in the medium containing leguminous plants using tumor-bearing animal model, and to elucidate the mechanism of anti-hepatoma activity by gene expressions in tumor and reculture of hepatoma cells from tumor.
