2012-02-212024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/682554摘要:本年度主要目標在於建立重大病媒害蟲桃蚜藥劑篩檢之技術平台,期能達成:(1) 桃蚜卵巢體外培養系統之建立;(2) 桃蚜卵巢細胞體外培養系統之活性測試;(3) 桃蚜體外培養卵巢對藥效反應之測試;(4) 殺蟲藥劑對細胞活性與生殖基因表現之影響測試。藉由桃蚜卵巢體外培養系統之建立、細胞活性檢測技術之開發、藥效反應評估標準之檢驗,以及殺蟲藥劑對細胞活性與生殖力之影響評估等實驗,發展出一套快有效之藥劑檢平台。桃蚜體外培養系統之建立旨在掌握藥劑有效施用時段;藥效之檢測反應,可由核糖核酸之完整性與管家基因之表現程度,作為指標;在殺蟲藥劑測試方面,除了檢測藥劑對細胞活性及抗藥性基因之表現影響,更可運用於檢測藥劑是否對生殖基因產生抑制效能。我們期待利用分子工具定量基因表現之優勢,大幅提升蚜蟲藥劑篩檢之準確性與效能。<br> Abstract: We propose to establish an in vitro culture system for ovaries dissected from the green peach aphid Myzus persicae, aiming to apply molecular tools to detect cell viability, drug effects, and insecticide efficacy. Through integrating all of these techniques, we expect to establish an effective platform for screening chemicals with the potential to control the growth and proliferation of aphids. After the establishment of the in vitro culture system, we aim to identify the maximum period of cell viability so that we can perform drug treatment within such effective time window. Comparison of cell viability before/after the treatment of drugs is judged according to the integrity of rRNA and the expression level of housekeeping genes. Furthermore, we will detect the expression of insecticide-resistant genes as well as reproduction genes in the ovarioles subjected to various kinds of chemicals.
