2023-01-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/694460次世代基因定序技術 (next-generation sequencing; NGS) 可以偵測多種植物病原 ,排除偽陽性,並檢測未知病原及發現潛在風險。傳統高通量 NGS 價格昂貴,本計 畫目標是結合國立中興大學與國立臺灣大學團隊,進行經濟快速型通量次世代基因 定序病害檢疫平台建立,測試以低成本、快速方式達到精準病害檢測之可能性,以 期未來應用於檢疫業務。112年度計畫目標包含:1. 針對檢測樣本最大化進行測試 ;2. 建立植物檢疫病毒資料庫;3. 以NGS定序技術解序分析進出口花卉及其種苗病 株之病毒病害種類,建立有害生物資料集供後續比對及風險評估分析。藉由測試建 構此經濟快速型通量次世代基因定序病害檢疫平台及專屬進出口花卉種苗病毒病害 資料集,以次世代基因定序技術應用於輸入植物檢疫之潛力,未來倘完成平台建立 ,可有效掌握我國及進口植物產品潛在的病毒病害種類,掌控我國防檢疫主導權 ,提升病害檢測量能。 The next-generation sequencing (NGS) can detect multiple plant pests, exclude false positives, and identify unknown pathogens. The cost for traditional NGS is still expensive; thus, cost down is the critical point for the NGS applied on border quarantine. This proposal combines the effects of National Chung-Hsing University and National Taiwan University to cooperate on testing the possibility of fast and economic throughput NGS diagnosis of plant viruses, for future application on plant quarantine. In the year 2023, we are planning to accomplish: 1. To maximize the detection of small molecule nucleic acid samples. 2. collection of the critical plant quarantine virus reference sequences, and 3. Using NGS sequencing technology to desequence and analyze the types of virus diseases of imported and exported flowers and seedlings, and establish a pest data set for subsequent comparison and risk assessment analysis. This project can explore the potential to build up a fast, economic throughput NGS technique and a friendly platform for plant virus detection and simplifying the NGS cDNA sample preparation procedure.
