2013-08-012024-05-13https://scholars.lib.ntu.edu.tw/handle/123456789/643043摘要:本研究將延續本實驗室在體染色體常用 STR基因上尋找連鎖之 STR基因,以配對形成高鑑別力之組合,並解決究係突變或為無親緣關係之親緣鑑定問題。本研究將尋找X染色體之連鎖 STR基因,建立 X-STR單倍型系統,解決具 X染色體關連性之親緣鑑定問題。Y染色體之 STR基因均不參與聯會,其組合形成的單倍型可成為父系親緣之重要標記。而目前發表之 X染色體之 STR基因大多相互獨立不連鎖,因此難以形成親緣標記,在法醫親緣鑑定上功能有限。本研究將建立相互連鎖之X-STR基因,建立X-STR單倍型之親緣標記,一則提高人別鑑定之鑑別力,一則解決具 X染色體關連性之親緣鑑定。本研究將進行三年,第一與第二年將從 X染色體 DNA序列中找出兩組各在連續 100萬鹼基內含有 10個可能的 STR基因,設計 PCR引子、進行 PCR複製與毛細管電泳分析,評估多型性,並建立多型變化的序列結構。第三年將此兩組連鎖之 X-STR基因組合為多重 X-STR基因螢光毛細管電泳分析系統,分別在兩組 PCR複製反應完成後,即可進行毛細管電泳分析,以達到快速分析之效果,並進行族群分析,建立基因頻率與連鎖基因組成單倍型的頻率、分析 STR基因的連鎖關係及與現有 X-STR基因間的連鎖關係等。三年完成後,本計畫不僅建立可增加人別鑑定能力的 20個 X-STR基因,並可解決具 X染色體關連性之親緣鑑定。<br> Abstract: This project will extend this lab’s study on searching the linked autosome STR loci commonly used in forensic lab, forming a highly discriminatory power of paired STR and solving the problem of mutation or unrelated in the mismatch case of parentage testing. This project will search the linked X-STR loci and combine as a X-STR haplotyping system. This can be used in X-related kinship test. Since Y-STRs are not cross-over in meiosis, Y-STR haplotype can become a marker of paternity. Most X-STRs, however, are not linked, they cannot become family markers and provide enough information in kinship test. The haplotype of linked X-STRs established by this project can become family markers. They can provide highly discriminatory power either in personal identification or in X-related kinship test. The study will take 3 years. In the first and second years, this study will isolate 2 sets of each with 10 new X-STR markers which locate in X chromosome within 100 Mb range. Evaluating the polymorphism by PCR technique and repeat DNA structure will be performed. In the third year, two multiplexs each with 10 new STR markers will be established in fluorescent capillary electrophoresis for fast screening. Population study on gene frequencies, haplotype frequencies and linkage analysis will be established.
