2014-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/711767摘要:申請人 102 年度國科會計畫,針對甘藍於高溫淹水、高溫不淹水、適溫淹水及適溫不淹水下之轉錄體進行定序,共有 214,357,592 條 reads,共組出83,910 條 contigs, N50 為 1,115 nt,組裝品質良好,reads 使用率為 95.4%。本計畫擬進一步分析上述高溫淹水反應相關之微型核酸及基因之表現情形,以完整建立甘藍在高溫淹水逆境下之轉錄體資料庫。第一年度擬分析比對甘藍於高溫淹水及常溫不淹水下之微型核酸 (micro RNA, miRNA),探討受高溫淹水逆境影響之微型核酸變化,找出甘藍之保留性與新的微型核酸,並搜尋受微型核酸調控之轉錄體。第二年度則擬找出在高溫淹水逆境下有差異表現之基因,利用Real-time PCR 進行個別確認,同時確認生理表現,並建立與高溫淹水相關之轉錄體基因組與代謝路徑資料。本計畫之執行將有助於了解甘藍高溫淹水生理,差異表現之基因組與微型核酸未來或可供做選育高溫淹水耐受品系分子標記之參考。<br> Abstract: In 2013 NSC project, trascriptomes in the leaves of cabbage under heat and waterlogging, heat, waterlogging and control were sequenced to search for the differentially expressed genes and the affected metabolic pathways. Totally 214,357,592 reads were obtained. Using de novo assembly with CLC software, 83,910 contigs were assembled with N50 being 1,115 nt. The usage of reads was 95.4%. The quality of assembly was good enough for subsequent transcriptome analysis. The aims of this project are to analyze the expression of micro RNA at the transcript level and the genes involving the responses to heat and waterlogging stresses to establish the database of comprehensive transcriptomes of cabbage. In the first year, micro RNAs expression in the leaves of the control and the cabbage under heat and waterlogging will be analyzed, to distinguish the conserved and novel micro RNAs and to find the part in transcriptome regulated by the micro RNAs. In the second year, differentially expressed genes under heat and waterlogging will be searched and confirmed with Real-time PCR to establish the gene sets and metabolic pathways involved. The execution of this project will be helpful to understand the physiology of heat and waterlogging in cabbage.
