2010-01-012024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/662681摘要:Potyviridae科Potyvirus屬病毒為長絲狀、單鏈正股RNA病毒,是植物病毒中最大的一屬,在田間由蚜蟲以非永續方式傳播,對農業造成相當大的損失。若能發展出一套可快速且成功檢測及鑑定各種potyvirus的方法,對於各國的農業以及植物防檢疫工作應有極大的助益。本計畫希望研發出可快速檢測potyvirus之ELISA和快檢試紙套組,以及以寡核苷酸晶片為基礎的potyvirus快速鑑定套組,以協助檢測各種植物樣品中是否有potyvirus存在,並鑑定其種類。本計畫今年將執行下列工作項目:(1)大量製備和純化potyvirus廣效性單株抗體與多株抗體,(2)決定potyvirus ELISA檢測套組的產品型式,(3) potyvirus快檢試紙之製備,(4)快檢試紙之改良與最佳化之調整,(5) potyvirus快速檢測套組產品的測試。所研發之potyvirus快速檢測套組包括ELISA和快檢試紙產品,可提供國內研究與防檢疫單位測試使用。<br> Abstract: The genus Potyvirus (in the family Potyviridae) contains the largest number of plant virus species which have single-stranded, positive-sense RNA genomes with filamentous particles. Potyviruses are transmitted by aphids in a non-persistent manner and cause significant losses in a wide range of crop plants. If the rapid detection and identification kits are developed for potyviruses, the agricultural industry and the works of plant inspection and quarantine will greatly benefit from them. The goals of this project are to develop the rapid detection kits including ELISA and immunostrip, and the rapid identification kits based on oligo chip for potyviruses. By using these kits, the existence of potyvirus in plant samples can be investigated and the virus species can also be confirmed. In this year the following works will be executed: (1) Large amount of broad-spectrum monoclonal and polyclonal antibodies will be prepared and purified. (2) The format of potyvirus ELISA kit will be determined. (3) Potyvirus immunostrip will be prepared. (4) The format of immunostrip will be modified and optimized. (5) The performance of potyvirus ELISA and immunostrip will be analyzed.
