2008-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/701115摘要:本研究的目的為(1)評估PCV2 對血液單核球所衍生不同分化成熟度的巨噬細胞之影響,(2)探討PCV2 引發肉芽腫性炎症反應的機制,(3)釐清巨噬細胞與淋巴細胞間的交互反應及免疫活化因子在PCV2 引發肉芽腫性炎症反應上所扮演的角色。;計畫中擬以活體外方式,經由豬隻周邊血液單核球所衍生之不同分化成熟度的巨噬細胞(monocyte-derived macrophages, MDMs)之誘發、定性及培養技術之建立,再於單純PCV2 接種或同時混合 有淋巴細胞及/或細菌內毒素(LPS)或T 淋巴細胞致裂原concannavalin A 等免疫活化因子培養條件下,分析相關免疫細胞的PCV2感受性、細胞存活率、細胞總數、多核巨細胞的形成、抗原呈獻、趨化能力、吞噬力、殺菌力、細胞凋零率、細胞毒殺能力,以及各類炎症反應中介質及細胞激素的產生能力等。<br> Abstract: The objectives of the present study are (1) to evaluate the effects of PCV2 on blood monocyte-derived macrophages (MDMs) of different maturation stages, (2) to elucidate the pathogenesis of PCV2-induced granulomatous inflammation, and (3) to determine the role of macrophage/lymphocyte interaction and immune-activating factors on the development of PCV2-induced granulomatous inflammation. MDMs of different maturation stages will be generated, characterized, and inoculated with PCV2 with or without co-culture with lymphocytes and/or lipopolysaccharide and concanavalin A in vitro.
