2017-01-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/660095摘要:近年來,食品安全事件頻繁發生,乳製品的安全議題倍受重視。台灣地處於亞熱帶氣候,環境以高溫多濕情況居多,常導致乳牛之乳產量下降與卵巢功能衰竭造成牛隻因繁殖障礙的因素大量淘汰牛隻,最終造成酪農經濟上莫大的損失與牛乳供應量的不足。近年來的研究中得知幹細胞具有自我更新和分化能力,可以取代受損的組織或促進修護受傷組織的能力,因此幹細胞療法已被作為疾病和再生醫學治療中重要之研究工具,實驗已經證實了他們移植後具有使損傷細胞再生和恢復組織功能的潛能。羊水幹細胞具有較高的自我更新能力;有研究顯示,人類羊水幹細胞能存活並增殖於卵巢早衰的小鼠,並且可能具有恢復卵巢功能的潛能;另外,本研究團隊中已證實,小鼠羊水幹細胞移植於化療引起卵巢早衰的小鼠,亦可恢復其部分卵巢之功能。但截至目前為止尚無任何研究發表牛羊水幹細胞治療牛隻卵巢衰竭的結果。基於此,本研究計劃,擬以荷蘭牛為模式,首先建立牛隻羊水來源幹細胞體外培養系統,進行細胞特性分析,建立判定卵巢衰竭牛隻的標準程序,並建立注射牛隻羊水幹細胞於萎縮卵巢牛隻之方法,進而完成牛隻羊水來源幹細胞對於卵巢衰竭牛隻之治療試驗。以上成果將提供牛隻羊水來源幹細胞之學術研究,並且有利於協助酪農解決現場牛隻卵巢衰竭的問題。<br> Abstract: Up today, food safety events occurred frequently and dairy product security issues are proposed. Taiwan is located at the subtropical climate, hot and humid environment. The weather results in decreased milk production and ovarian failure. This factor causes reproductive failure in cattle due to a large number of cattle culls. Eventually, it results in a great economic loss such as insufficient dairy production and milk supply. Research demonstrated that stem cells have the ability to selfrenewal and differentiation, moreover, it can replace damaged tissues and promote tissue repairing. Therefore, stem cell therapy has been used as the treatment of diseases and regenerative medicine. Experiments were confirmed that stem cells hold the promise to restore tissue function. Amniotic fluid stem cells have a higher self-renewal capacity with low immunogenicity. In recent years, amniotic fluid stem cells have been extensively studied in various tissues or organs of the damage repair in regenerative medicine, such as muscle, kidney, lung and heart. It is considered that amniotic fluid stem cells are potential source in regenerative medicine. Researches have shown that human amniotic fluid stem cells are capable of restoring chemotherapy-induced premature ovarian function. Up today, there is no any report illustrated that whether amniotic fluid stem cells hold the promise to cure cattle ovarian failure. Hence, this research project is to establish cattle amniotic fluid derived stem cells for cell and check the cell characterization and investigate the determination of ovarian failure cattle standard procedure. Cell transplantation approach is about to establish to investigate whether these amniotic fluid stem cells are capable of restore the cattle ovarian function.
