2013-08-012024-05-13https://scholars.lib.ntu.edu.tw/handle/123456789/650237摘要:背景及目的:本計畫希望找尋並研究適合運用於感應式溫熱療法的磁性材料。在人體的礦物質中有鐵及硫存在,而且需要經常攝取。根據相關文獻報導,在鐵硫化合物中廣為研究且具有磁性的物質為硫複鐵礦及磁黃鐵礦。本研究將以磁性硫複鐵礦為主要的磁性研究材料,實驗並找尋簡易的製作方式、適合的製作條件以及進行相關的生物實驗。方法及實施進度:計畫第一年(2011):對材料進行合成、磁性及體外生物相容性測試(LDH細胞毒性測試, MTT對細胞增生測試)。並進行體外熱治療試驗,測量在外加磁場下的腫瘤細胞以及正常細胞對溫熱治療的反應。不斷嘗試不同的參數,以找出最適合的磁場強度、交流電頻率。第二年(2012):進行一個月(subacute toxicity)及三個月(chronic toxicity)之體內生物相容性測試。建立癌症老鼠之動物模式,進行體內實驗,追蹤腫瘤大小,以實際瞭解我們的材料在生物體內治療癌症的效果。同年開始建立第三年(2013):根據磁場強度、熱療法頻率再去做更細的分類,以建立劑量—療效之曲線。目標是希望盡量減少熱治療之頻率。另將實驗動物模式繼續擴展成「骨骼癌症轉移」模式。第四年(2014):將對此種材料達成熱治療效果,進行深度之分子生物學機轉研究。<br> Abstract: Hyperthermia is one of the most popular research topics for cancer therapy. This research is aimed to develop a biodegradable magnetic nanoparticles based on Greigite for cancer hyperthermia. The greigite contains iron and sulfate, which we people digest every day. After applying external magnetic field by AC current, we can raise the temperature of the material. Therefore the material is a possible candidate for cancer hyperthermia therapy.Material and Methods :First Year(2011): We will perform the cytotoxicity tests, including MTT and LDH test. We’ll explore whether the cell adhesion, proliferation and differentiation being affected by the material itself without external magnetic field. The in vitro test with external magnetic field will be carried out. The thermal sensitivity of normal cells and cancer cells were measured.Second Year(2012): We will perform in vivo biocompatibility test for one month (subacute toxicity) and three months (chronic toxicity). We will establish an animal model and test the magnetic DCPD particles on mice with cancer.Third Year(2013): We will further classify the mice into different groups, in terms of the strength of magnetic field, the frequency of hyperthermia therapy and aim to reduce the frequency of therapy.
