2004-08-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/694494摘要:本研究計畫之目的為萃取與分離臺灣扁柏心材、樹皮、葉子、枝條等各部位抽出成分,評估其各種生物活性,包括抗真菌、抗細菌、抗蟎、抗病媒蚊、抗氧化、抗腫瘤等,並鑑定具生物活性成分之化學結構。首先,以溶劑萃取臺灣扁柏各部位之抽出物,續以不同極性之溶劑萃取區分為各分離部,並進行各項生物活性試驗,篩選出各種生物活性效果最佳之分離部,再經由層析技術加以分離與純化,所得之純化合物則利用質譜、紅外線光譜及核磁共振光譜等儀器分析加以鑑定化學結構,同時亦將純化合物進行各項試驗以便確認其生物活性,甚至利用化學改質方法增強其生物活性;另外,亦將進一步解析純化合物對不同生物或腫瘤細胞之抑制機制。本計畫如能順利完成,將可了解臺灣扁柏抽出物的各項生物活性,並使具有生物活性之成分應用於保健及醫療等用途,進而達到擴展<br> Abstract: The objectives of this project are to evaluate the bioactivities of Chamaecyparis obtuse var. formosana extracts and to isolate the bioactive pure compounds. At the beginning, different tissues (heartwood, bark, leaf, and twig) of C. obtuse var. formosana will be extracted with various organic solvents. The extracts are then separated into several subfractions by liquid-liquid partitions. To obtain the best active subfractions, the subfractions collected are under the examination for various kinds of assays, including antifungal, antibacterial, antimitic, mosquito larvicidal, antioxidant, and antitumor bioactivities. The pure compounds existed in the subfractions are finally isolated by chromatographic separation techniques and identified by modern spectroscopic analyses, such as MS, FTIR, NMR etc. Identified compounds are also examined with bioactive assays to confirm their bioactivities. Furthermore, we will try to enhance the bioactive effectiveness of compounds identified by using chemical modifications. In addition, the mechanisms for inducing bioactivities or apoptosis in tumor cells will also be elucidated.
