Effectiveness and safety evaluation require thorough investigation prior to apply on human patients. Compared with commonly used murine models, considering organ size, anatomical and physiological aspects, porcine model is more closely related to human patients, and is therefore a suitable animal model for conducting cell therapy evaluations. No current porcine model has been established for peritoneal injury and autologous mesothelial cell therapy evaluations, by establishing porcine model, we will be able to accelerate testing, evaluation processes and clinical applications for (autologous) mesothelial cell therapy in uremic patients undergo peritoneal dialysis. This subproject aim to establish porcine model to evaluate the effectiveness, safety and tumorigenesis potency of mesothelial cell therapy, and will be conducted at the veterinary school and animal teaching hospital at NTU main campus by an experienced team (including 3 licensed veterinarians, pathologist and surgeon). Peritoneal injury will be induced with 0.05% sodium hypochlorite as the method described previously in rodent species. The success of peritoneal injury model will be evaluated by histological examination including the lining of mesothelium, the thickness of submesothelial compact zone, vasculopathy, inflammatory cell infiltration, and tumor formation.
