2009-03-012024-05-13https://scholars.lib.ntu.edu.tw/handle/123456789/655178摘要:國際上已有一些相關研究發現,不過國內適用的分子標記仍未確認,所以本計畫的目標是針對繁殖性能的分子標記進行研發。而近年來與母豬繁殖相關性狀的候選基因分析,國際上報導的很多,包括動情素受體、助孕素受體與瘦身素受體、造骨蛋白(osteopontin)等基因,都被認為是影響繁殖性狀的重要候選基因。藉由這些在候選基因母猪繁殖性能為影響養猪產業生產效率之重要因子,本研究團隊自製24000基因晶片並用以檢測桑椹胚發育至早期囊胚階段中,mRNA表現量顯著增加或降低之基因作為候選基因,選殖到10個候選基因的調控區域,並分析其單核苷酸多型性(SNP) 分子遺傳標記,同時利用大量鑑定SNP 基因型技術以瞭解其與窩仔數性狀關係。經由30 頭純種藍瑞斯母猪定序後發現,在10個候選基因裡,共發現59個新的SNP。利用SNP基因定型系統鑑定161頭藍瑞斯母猪之SNP基因型和基因型對於母猪之出生仔猪總頭數與存活仔猪頭數之差異。結果發現,共有三個基因7個SNP與藍瑞斯品種母猪之出生仔猪總頭數有顯著關係存在。目前已發現的三個與產仔數有關的基因分子標記,正在申請專利中,相信這些在臺灣開發的分子標記,最能適用於促進臺灣的養豬產業,本計畫擬開發成檢測平台與套組以發揮產業化的目標與增進養豬產業發展的效用。 <br> Abstract: Reproduction performance of sows is the most important factor for pig industry. We have made a gene chip for 24000 genes specifically for pig research. Using such gene chips, we found many genes specifically expressed in certain stages during embryonic development. We cloned promoter sequences of 10 genes and found their polymorphism by sequencing these genes of 30 Landrace pigs. We also found 59 new single nucleotide polymorphisms from these genes. Three of them are significantly correlated to reproduction performance of sows. We have filed for patents.
