2019-01-012024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/664454摘要:紅豆 (Vigna angularis) 富含多酚類、皂&#33527;及膳食纖維,在先前的研究中發現紅豆萃取物具有保護心血管、抗高血壓及改善胰島素阻等功效。實驗室先前研究也證實紅豆水萃物具有抗肥胖之功效,為解決紅豆在採收季節發生滯銷,無法長久存放,及格外品的處理等問題,除了促成企業團購及推廣外,政府也積極與業界加工廠及學術機構合作,以推出各式加工品或以提高紅豆價值的方式幫助農民。非酒精性脂肪性肝疾病 (NAFLD) 為目前被全世界重視的主流公共衛生問題,其發病機制與基因,環境和營養息息相關。已知 NAFLD 與肝臟的發病進程有關,從肝臟脂肪變性到肝硬化和非酒精性脂肪肝炎 (NASH),甚至 NAFLD的一個亞型可以進展到肝癌。直至今日,如何預防和治療肝硬化、脂肪肝和慢性肝炎等常見肝臟疾病仍然存在許多問題。因此,實驗設計以 HepG2 人類肝癌細胞添加油酸誘導,模擬非酒精性脂肪肝環 境下的肝細胞模式進行評估,更進一步利用高脂飲食 (high fat diet, HFD) 促進之小鼠非酒精性脂肪肝模式探討國產高雄9號紅豆水萃物之化學預防效果及分子機制,並進行活性成分分析及產品開發。期望藉由此計畫找出國產紅豆多樣性利用價值,評估紅豆水萃取物在 NAFLD 化學預防功效與分子機制,這些結果將有助於台灣雜糧作物進一步的新穎性發展。<br> Abstract: Adzuki bean ( Vigna angularis ), an important economic crop of Taiwan, are known as containing various polyphenols, saponins and rich in dietary fiber. The extracts of adzuki bean have been found to show positive effect on cardiovascular protection, anti-hypertenison and improvement in insulin resistance. Previously, our laboratory has found that Adzuki bean water extract could demonstrate anti-obesity effect. In order to solve the problem of slow sales, long-term storage and handling defective or secondary products during harvesting season, active co-operation of government with processing industry and academic institution has recently become a successful problem solving strategy to improve the economic value of Adzuki bean by launching various processing or functional products, besides of facilitating group purchases or promotion. Nonalcoholic fatty-liver disease (NAFLD) is now attached to the importance of mainstream public health concern worldwide. Its pathogenesis mechanism is closely related to genetic, environmental and nutritional issues. Besides, it has been noted that lipid steatosis in liver may lead to cirrhosis and non-alcoholic steatohepatitis (NASH) and therefore, NAFLD is known to be highly related to leading process of liver diseases. Moreover, NAFLD has also been reported to be a potential developing stage in liver cancer progression. However, there are still some unclarified problems in prevention and treatment of liver diseases like liver cirrhosis, fatty liver and chronic hepatitis. Therefore, the experimental project designed to investigate the possibility of additional value of domestic adzuki bean, by evaluating effect of adzuki bean water extract on chemoprevention of NAFLD and clarifying its possible molecular mechanism. In fatty liver cell model, by stimulating HepG2 human liver cancer cell oleic acid, it is possible to imitate environmental condition of NAFLD under cell model and evaluate the preventive effect of Adzuki bean water extracts in fatty acid uptake. Furthermore, designed animal model aims to study the chemo-preventive effect and molecular mechanism of adzuki bean on hepatic streatosis promoted by high fat diet (HFD).
