2011-11-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/687952摘要:番茄是茄科植物,為世界重要蔬菜作物,可供鮮食與加工用。為提高番茄果實之產量與品質,以及避免颱風、豪雨、病蟲害等危害,目前臺灣農民大多採用設施栽培。但因農民過度施用肥料,加上設施栽培缺乏雨水淋洗,不僅提高生產成本,也造成土壤鹽類的累積。合理化施肥為解決上述問題之一重要對策。為達合理化施肥之目標,即需檢測植株營養狀態。然而目前大多採用植體或土壤樣品進行生化方法的檢測,耗時且需一定的設備及技術。再者,以磷來說,即使土壤中檢測出大量磷,卻僅有少部分可被植物吸收利用,導致缺磷為一常見現象。磷在番茄生長初期及開花階段扮演重要角色。因此本研究期望利用農桿菌注入法短暫表現的系統,建立方便、快速檢測番茄植株體內磷酸鹽含量是否充足的方法,以達合理化施肥、減少肥料支出、並維護生態環境之目的。為達上 述目標,本二年期研究計畫內容包含有(一)篩選專一且敏感表達番茄磷酸鹽含量不足的基因(二)啟動子與報導基因的基因建構(三)建立番茄農桿菌短暫表現的系統。未來更可進一步將此技術應用於其他作物及診斷不同種類的營養元素。<br> Abstract: Tomato (Solanum lycopersicum) which belongs to Solanaceae is an important vegetable crop in the world for fresh and processing consumption. In order to increase fruit yield and improve fruit quality, and avoid injury from typhoon, torrential rain, pest and disease, most farmers in Taiwan apply greenhouse cultivation for growing tomato. Because over-fertilization by farmers and lacks leaching from rain of greenhouse cultivation, it often leads to increase cost and accumulation of salt in the soil. Reasonable fertilization is one of important approach for solving above problems. To achieve the purpose of reasonable fertilization, it is essential to measure the nutritional status in plants. However, the commonly used element analysis of soil and plant material to assess fertilizer requirement is time-consuming and requires expensive equipments and high techniques. Furthermore, in the case of phosphorus, high amount of phosphorus detected in the soil is usually unavailable for plants, resulting in deficiency. Phosphorus plays important roles in initial growth and flowering stage of tomato. To achieve the goals of reasonable fertilization, decreased cost of fertilizer and protection of ecological environment, we will employ an agroinfiltration transient expression system to develop an in planta method for conveniently and quickly assay of phosphate (Pi) status in tomato plant. For above goals, the following studies will be taken within two years. (1) Screening Pi starvation-induced genes which can properly and sensitively respond to Pi status in tomato. (2) Construction the promoter: reporter constructs. (3) Set up the agroinfiltration system in tomato. (4) Greenhouse test.
