2005-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/710763摘要:金線連為台灣民間珍貴的食藥用植物,被相信具有鎮痛、降血糖、降血壓、護肝、利尿與消炎作用,其生物刺激、生理與藥理的活性已經被證實。其對免疫能力的調節已被確認,並歸因於其所含之多醣。本研究群的初步試驗顯示,其水溶性多醣可以刺激人類週邊血液單核球分泌腫瘤致死因子(TNF-α), 間白素-6 (IL-6)的分泌,但對間白素-2 (IL-2) 則無影響。本計畫擬以三年時間分離、純化金線連具活性的多醣,並對其分子結構特徵與活性與結構的相關性進行研究。第一年的工作已建立活性多醣之分離、純化方法並分析其理化性質與細胞免疫調節活性。第二年工作將分析中性與酸性活性多醣的化學結構包括單糖組成、鍵結位置、組態與分子量分佈。第三年工作重點為利用修飾多醣分子研究分子結構與細胞免疫活性刺激能力之析關性並對中性與酸性多醣的相乘性進行研究。<br> Abstract: Anoectochilus formosanus is a unique species and valuable folk medicine used as an illness remedy in Taiwan. The herb has used to treat hypertension, diabetes, consumption, nephritis, and liver diseases in alternative medicine. The bioactivity, physiological function and pharmacological effect have been confirmed. The immune-modulating activities of the extracts of whole plant have been confirmed by recent studies. The activities have been associated with its polysaccharides. According our preliminary results, the water-soluble polysaccharides of the whole-plant significantly stimulated human peripheral blood mononuclear cells to produce tumor necrosis factor-α, TNF-α, and interleukin-6, IL-6, but not IL-2. This 3-year project is planning to isolate the bioactive polysaccharides from the plant, to elucidate their structural characteristics, and to investigate the molecular structure and bioactivity relationship. The tentative schedule has been set as followings: First year, the works focus on the protocol to isolate and purify the bioactive fractions and confirm their cell immuno-modulating activity and chemical and physical properties. The works of second year focus on the structural feature of neutral and acidic bioactive polysaccharides. It is the goal to establish the data of sugar composition, glycosidic linkage type, anomeric configuration and molecular weight and distribution. Structural modification of bioactive polysaccharides will conduct on the third year to study the relationship of molecular structure and its bioactivity.
