2007-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/699881摘要:本研究旨在將已帶有纖維素分解&#37238;(cellulase)基因及植酸分解&#37238;(phytase)基因等之轉基因小鼠與轉基因豬,進一步針對轉殖基因在胰臟中的轉錄轉譯調控機制,及其在性腺傳承之效率與轉基因蛋白質產物對於飼料中纖維素及有機磷消化分解能力等,分別進行深入詳實之研究外,並擬配合纖維素結合蛋白質(cellulose binding protein, CBP)可以顯著提高纖維素分解&#37238;對飼料中纖維素結合能力之特性,試圖將該基因之全長cDNA與豬胰澱粉&#37238;基因啟動子(porcine pancreatic amylase promoter)及釋泌信息&#32957;(signal peptide)進行構築成轉殖基因(transgenes),俾供後續用以產製帶有該外源基因之轉基因小鼠與轉基因豬,冀能使豬隻對於飼糧中纖維素之分解消化能力,更形提昇之效果。 此外,研究亦將持續進行產製攜帶有木聚醣分解&#37238;(xylanase)基因、葡聚醣分解&#37238;(glucanase)基因之轉基因動物,並藉由攜帶有多種消化性分解&#37238;之轉基因動物提供更多營養學上之分析研究,預期將可達到改進家畜對飼料中營養分的消化與利用之目的,並增加生長效率、減少排泄物,進而達到環保與提升農民收益之最終目標。 至此,本研究提供了國內畜牧產業發展的新方向,並予生物技術工程一全新之應用領域,唯有對轉基因動物之營養相關試驗有更精進之研究,始可對此相關發明有更進一步利用及推廣。 <br> Abstract: The objective of this study was already to generate transgenic animals carrying designed transgenes animals which includes a porcine pancreatic amylase promoter and cellulase and phytase gene. further to the transcription and translation the regulation and control mechanism in the pancreas, and efficiency gremlin transmission and transgenic protein digest the ability of resolving etc. to the cellulose and organic phosphorus in fodder, outside deepenning full and accurate research separately, we plan to cooperate with the cellulose binding protein(CBP)that can improve the cellulose combine characteristic of ability to fodder, attempt total length cDNA and porcine pancreatic amylase promoter And clear up the information of secreting signal peptide Construct and transfer to the transgenes ,In order to follow-up of supporting being in order to is it make have other source transgenic mice and pig to produce, Ji can make pig for raise cellulosic digestion power of resolving in the grain, appear result that promote even more only.
