2010-08-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/688512摘要:酚系抗氧化物質,為自然界蔬果中主要的抗氧化物質,可以減少心血管疾病的發生;另外,它也可以藉由疏水基鍵結和氫鍵鍵結方式抑制某些酵素活性,因此發現熱水沖泡烏龍茶中的多種酚類物質對胰臟脂解酶具有抑制作用,具有減重效果。農委員會(2007)指出:荔枝年產量約90,000公噸,產值15億元。由於未授粉荔枝花朵會掉落至地面成為廢棄物,而荔枝水萃液中發現含有大量酚系抗氧化物質。 計畫申請人預計花3年時間,首先分析並判定荔枝花水萃液中主要的酚系抗氧化物質與含量,再探討其3種保健功能:1)降血脂:探討荔枝花水萃液是否可以調控肝臟內LDL receptor,膽固醇及三酸甘油酯生合成酵素,與增加膽固醇代謝/排泄?2)減緩非酒精性脂肪肝形成:飲用荔枝花水萃液是否改善高油脂/膽固醇飲食下導致的脂肪肝形成,並調查其肝細胞氧化受損情形、MMP-2/9、TIMPS、Hsp70、細胞凋亡(apoptosis)訊息路徑是否有影響?3)減緩肥胖症形成:飲用荔枝花水萃液是否可以在高熱量飲食下有效降低體脂肪形成,而其機制分成3方面探討:外源性三酸甘油酯及膽固醇的吸收;內源性三酸甘油酯及膽固醇的生合成;體內脂肪與膽固醇的代謝與消耗? 預預預預期了解荔枝花水萃液中主要的酚系抗氧化物質與含量,並藉由動物模式下初步探討其降血脂、護肝、減重之保健功能,並藉由此結果提供日後繼續探討其他保健功能之依據。<br> Abstract: Litchi (Litchi chinensis Sonn.) is an economic fruit in Taiwan, which is valued of NT$ 15 billion per year. Litchi-flower-water-extract (LFWE) contains plenty of antioxidative phytochemicals, i.e. phenols, flavonoids, and tannins, which own several health properties, i.e. cardioprotective, antiobesity, and antitumor effects. First, the principle investigator will do a completely chemical analysis of antioxidative phytochemicals of LFWE. Then, three health functionalities of LFWE: 1) hypolipidemic effect, 2) amelioration of a nonalcoholic fatty liver (NAFL) induced by a high fat/cholesterol diet, and 3) antiobesity effect will then be dug into. In the animal trial, major analyses will include 1) serum lipids, i.e. triglyceride, cholesterol, and HDL-C concentrations, as well as the cardiac index (TC/HDL-C), 2) hepatic and fecal triglyceride and cholesterol, as well as fecal bile acid, 3) safety evaluations, i.e. blood pressure/sugar, liver function indexes (GPT, GOT, ALP, and γ-GTP), kidney functional indexes (BUN, creatinine, and uric acid), ketone body, and electrolytes (Na and K), 4) enzymes or proteins related to absorption of exogenous lipids, metabolism of endogenous lipids, and expenditure of lipids in body, 5) oxidative stress: MDA and GSH contents, as well as activities of GSH-Px, catalase, and superoxide dismutase in the body, 6) activities of hepatic damage protein and enzymes, i.e. TNF-α, MMP-2/MMP-9, and tissue inhibitor of metalloproteinases (TIMPS) in the liver, 7) Hsp70 and other signals of apoptosis in hepatic cells, 8) levels of hepatic cell death: % DNA fragments and TUNEL assay; 8) histological assessment of livers and epididymal fat. Via this three-year study, the ameliorations of LFWE on the symptoms of hyperlipidemia, NAFL, and obesity can be clearly investigated and understood.
